增殖细胞核抗原与卵巢癌细胞系化学敏感性 被引量:2

Relationship between PCNA expression and chemosensitivity of human epithelial ovarian cancer cell lines to anticancer drugs

摘要 目的 :探讨增殖细胞核抗原 (PCNA)与上皮性卵巢癌细胞系化学敏感性的关系。方法 :使用顺铂作用于A2 780及AD6 0细胞株。在MTT法测定耐药倍数基础上 ,用ABC检测PCNA表达。结果 :MTT测定AD6 0的耐药倍数为 2 .4。A2 780PCNA的表达低于AD6 0 ,且随药物浓度的递增染色较耐药组明显变浅。A2 780、AD6 0相同药物浓度作用后PCNA的表达有显著性差异 (P 0 .0 1) ;同一细胞株不同药物浓度作用后PCNA表达有显著性差异 (P 0 .0 1)。结论 :增殖细胞核抗原表达与上皮性卵巢癌细胞对细胞毒性药物的化学敏感性呈负相关。 Objective To learn the relationship between PCNA expression and chemosensitivity to epithelial ovarian cancer cell lines. Methods The human epithelial ovarian cancer drug sensitive cell lines A2780 and drug resistance cell lines AD60 were treated with cisplatin, respectively . MTT was used to measure the multiple of drug resistance . PCNA expression was detected by avidin biotin peroxidase complex method(ABC). Results The multiple of drug resistance of AD60 was 2.4. In A2780 group, PCNA expression was lower than that in AD60 group and the staining became gradually lighter with the increasing cisplatin concentrations. In PCNA expression, there was significant difference between two cell lines at the same cisplatin concentration( P ?0.001). And there were significant differences among PCNA expressions in the same cell line at differencent cisplatin concentrations( P <0.001). Conlcusion The present study suggested that PCNA expression was negatively related to the chemosensitivity of epithelial ovarian cancer cell lines to cytotoxic drugs.
