醛糖还原酶抑制剂对实验性糖尿病周围神经病大鼠内源性NGF及神经肽的影响 被引量:4

The influence of ARI on endogenous NGF and neuropeptides in rats of experimental diabitic neuropathy(DNP)

摘要 目的 探讨 NGF在 DNP发病中的作用和 ARI改善 DNP的作用机制。方法 用 STZ制成大鼠DNP模型后给予 ARI治疗 ,应用免疫组化和 elisa测量 DRG中 SP、CGRP和血清及坐骨神经 NGF含量 ,并与未治疗的 DNP模型和正常对照相比较。结果 实验性 STZ DNP大鼠 DRG中 SP、CGRP和血清及坐骨神经中 NGF水平下降 (P<0 .0 5) ,ARI治疗后均得到显著增高 (P<0 .0 5)。结论  NGF的产生和 /或轴突运输下降及其继发的神经肽缺乏在 DNP中起作用。 ARI可促进 Objective To investigate the role of nerve growth factor( NG F) in the pathogenesis of DNP and the effect of ARI on DNP.Methods DNP rats were treated with ARI.The content of SP and CGRP in dorsal root ga nglionand (DRG) the levels of NGF in serum and sural nerve were measured with im munohistochemistry and compared with those of untreated DNP rats and healthy co ntrols. Results The contents of SP and CGRP in DRG and the levels of NG F in serum and sural nerve of DNP rats decreased significantly(P<0.05).The treatment with ARI significantly increased them(P<0.05).Conclusio n The decrease of the production and/or axonal transport of NGF and the following decrease of neuropeptides contribute to DNP.ARI can promote the production of endogenous NGF.
