摘要
目的:研究钙通道拮抗剂尼莫地平对增殖性视网膜病变(OIR)的治疗作用及其与血小板衍生生长因子(PDGF)的相互作用。方法:以高浓度氧气诱导SD幼鼠 OIR模型。将P_2(出生后第2天)SD幼鼠随机分为8组(n=10),A为正常对照组,于自然空气中饲养,B为单纯OIR组,C、D、E为球后注射药物组,F、G、H为腹腔内注射药物组,后7组动物均于高浓度氧环境下饲养。球后及腹腔内注射组分别注射不同剂量的尼莫地平。取幼鼠眼球作普通病理切片及免疫组织化学检测,分别检测视网膜新生血管芽内皮细胞数目及PDGF的表达。结果:单纯OIR组视网膜新生血管芽内皮细胞数目及 PDGF的表达较正常对照组明显增加(P<0.01),球后注药大、中剂量组较单纯OIR组均明显减少(P<0.01),小剂量组则无明显变化。腹腔内注射各剂量组较未用药组均明显减少(P<0.01)。结论:PDGF在OIR组织中表达明显增加,而钙通道拮抗剂尼莫地平可抑制OIR的发生,并在某种程度上抑制PDGF的表达。
Objective: To study the therapeutic effect of calcium channel antagonist nimodipine on proliferative retinopa-thy(OlR) and its interaction with platelet-derived growth factor (PDGF). Methods: The model of hyperoxia-induced OIR was established in newborn Sprague-Dawley (SD) rats. SD rats P2(2 d after birth) were randomized into 8 groups and were designed as follows: Group A (normal control group) was bred in room air. Group B was pure OIR group. Group C, D, E were designed as retrobulbar injection groups. Group F, G, H were designed as intra-abdominal injection groups. Rats in group B-H were all exposed to a hyperoxia environment. Rats from retrobulbar injection groups and intra-abdominal injection groups were injected with nimodipine of different doses. Both eyeballs of newborn rats were enucleated for making common pathological sections and were examined by immunohistochemistory to count the nuclei of proliferative retinal vessels and to investigate the expression of PDGF in retina. Results: The number of nuclei of proliferative retinal vessels and the expression of PDGF in the pure OIR group increased significantly compared with those of the normal control group(P<0. 01), and decreased greatly in the high dosage and medium dosage retrobulbar injection groups compared with those of the pure OIR group(P<0. 01), while there was no great decrease in the low dosage group. In all the groups of intra-abdominal injection the decrease of the PDGF expression was significant (P<0. 01). Conclusion:PDGF expression is obviously increased in OIR patients) nimodipine can prevent OIR and it can inhibit the expression of PDGF to a certain extent.
