摘要
目的:观察妊高征患者胎盘组织细胞信号及传递蛋白相关基因表达的变化。方法:TRIzol法抽提6例妊高征患者和5例正常妊娠产妇胎盘组织总RNA并纯化mRNA;应用cDNA阵列检测与细胞信号和传递蛋白相关的1651个基因表达变化,信号扫描仪分析结果。结果:在1651条与细胞信号和传递蛋白相关基因中,妊高征胎盘组织与正常妊娠胎盘组织之间存在差异表达基因32个,包括上调基因26个,下调基因6个,其中已知基因31个,1个为未知基因EST片段。一些与介导免疫、水钠调节、细胞生长相关的基因表达上调。下调基因中也涉及了与免疫、水钠调节相关的基因。结论:妊高征的发病机制可能涉及了一些细胞信号和传递蛋白相关基因表达的变化。
Objective:To search for the differently-expressed genes in placental tissues in pregnancy induced-hyperten-sion(PIH). Methods: The total RNAs were extracted from the tissues by TRIzol and mRNA was purified by Midi Kit. The expression of 1 651 genes associated with cellular signal and transferring proteins were evaluated by cDNA array technique. Results:Totally 32 differently-expressed genes were identified in placental tissue of PIH. Twenty-six were up-regulated and 6 down-regulated. There was 1 EST among the up-regulated genes. The up-regulated genes included some genes associated with immune mediator, water and sodium regulation and the cell growth. Among the down-regulated genes, there were genes associated with immune mediator, water and sodium regulation. Conclusion: Some genes associated with cellular signal and transferring proteins are involved in the pathogenesis of PIH.
