非小细胞肺癌中hRAD17基因表达与淋巴结转移的关系 被引量:1

Overexpression of hRAD17 gene in non-small cell lung cancer correlated with lymph node metastasis

摘要 目的 研究非小细胞肺癌 (NSCLC)中hRAD17基因的表达频率及其与淋巴结转移的关系。方法 应用逆转录 多聚酶链反应 (RT PCR)方法检测了 5 0例肺癌标本的hRAD17表达。以人NSCLC细胞株SPC A1作为阳性对照 ,癌周正常肺组织作为阴性对照。结果  5 0例肺癌中 38例hRAD17阳性表达 ,阳性率为 76 %。 38例阳性病例中有纵隔淋巴结转移者为 31例(81.5 % ) ,而 12例阴性表达病例中仅 3例 (2 5 % )有纵隔淋巴结转移 ,两组间差异具有统计学显著意义 (P <0 .0 1)。结论 hRAD17基因在肺癌中高表达 ,其阳性表达与淋巴结转移密切相关。 Objective To investigate the frequency of hRAD17 gene expression in non small cell lung cancer (NSCLC) and its relationship with lymph nodes metastasis. Methods The expression of hRAD17 messenger RNA was evaluated by reverse transcriptase-polymerase chain reaction (RT-PCR) in 50 cases of surgical specimen of NSCLC tumors. The adjacent normal lung tissues were used as negative control, while a human lung cancer cell line SPC-A1 used as positive control. Results The hRAD17 transcripts were detected in 38(76%) of the tumor sample, among which 31 (81.5%) cases had node metastasis. On the other hand, 3 of 12 (25%) cases without hRAD17 overexpression were node positive. The expression of hRAD17 mRNA was correlated with lymph node metastasis (P<0.01). Conclusion The hRAD17 messenger RNA is frequently detected in NSCLC and its expression is correlated with lymph node metastasis.
