Effects Of Astragalus Membranaceus Injection On The Activities Of ATP ase In Caudal Epididymides Of Cd-Induced Rats

摘要 目的 观察黄芪注射液拮抗氯化镉诱导大鼠精子运动的毒性及对附睾尾组织ATP酶的影响。方法 将 48只SpragueDawley雄性大鼠随机分成 4组 :低浓度黄芪组 (A1 )、高浓度黄芪组 (A2 )、氯化镉组 (Cd)和对照组 (C)。预先连续 7d分别给A1 组和A2 组大鼠腹腔注射黄芪注射液 5g/ (kg·d)和 10g/ (kg·d) ,Cd组大鼠腹腔注射等量蒸馏水作对照 ,之后连续 7d给A1 组、A2 组和Cd组大鼠分别同时腹腔注射氯化镉溶液 [0 .4mg/ (kg·d) ]。染镉后第 8d处死大鼠 ,用计算机辅助精子分析系统 (CASA)分析附睾尾精子运动参数 ,并检测附睾尾组织ATP酶活性。结果 A2组精子活率 [( 4 3 .3 9± 5 .3 1) %]、前向运动精子百分率 [( 3 2 .3 3± 8.3 2 ) %]、曲线速度 [( 10 6.88± 2 8.5 1) μm/s]、平均路径速度 [( 5 8.0 3± 7.3 ) μm/s]和精子头摆幅度 [( 2 9.76± 3 .81) μm/s]等参数明显高于Cd组 [( 2 5 .83± 4.43 ) %、( 2 0 .92± 3 .0 4) %、( 90 .3 1± 12 .5 5 ) μm/s、( 3 6.91± 3 .2 6) μm/s和 ( 2 1.5 0± 3 .5 2 ) μm/s] (P <0 .0 5或P <0 .0 1) ;A2 组附睾尾组织Na+ K+ -ATP酶 [( 2 .0 3± 0 .2 8)× 10 - 3μmolPi/mgprot/hour]、Ca2 + -ATP酶 [( 2 .2 0± 0 .15 )× 10 - 3μmolPi/mgprot/hour]和Ca2 + Objective The effects of Astagalus membranaceus on Cd-induced rats sperm motility toxicity and the activities of ATPase in caudal epididymides were studied.Methods 48 adult male Sprague Dawley rats were divided into four groups:low concentration Astragalus membranaceus group(A 1),high concentration Astragalus membranaceus group(A 2),cadmium chloride group(Cd)and control group(C).Astragalus membranaceus injections[5g/(kg·d)and 10g/(kg·d)]were administrated by intraperitoneal injection to the A 1andA 2group rats.After 7 days,cadmium chloride(0.4mg kg·d was administrated by intraperitoneal injection to the A 1,A 2 and Cd group rats at the same time.The rats were sacrificed and observed sperms were isolated from the caudal epididymides of rats with diffusion method after the rats were induced by cadmium chloride at the eighth day. The effects of Astragalus membranaceus on the motility of isolated sperms of rats were observed by computer-assisted sperm analysis(CASA) system.Activities of ATPase were determined.Results The motility parameters of sperms (Mot,Percentage of a+b,VCL,VAP,ALH)in high concentration Astragalus membranaceus injection group[(43.39±5.31)%,(32.33±8.32)%,(106.88±28.51)μm/s,(58.03±7.3)μm/s and (29.76±3.81)μm/s])increased significantly compared with those of Cd-induced group[(25.83±4.43)%,(20.92±3.04)%,(90.31±12.55)μm/s,(36.91±3.26)μm/s and (21.50±3.52)μm/s])(P<0.05 or P<0.01).The activities of Na +K +-ATPase[(2.03±0.28)×10 -3 μmolPi/mgprot/hour]、Ca 2+ -ATPase[(2.20±0.15)×10 -3 μmolPi/mgprot/hour],and Ca 2+ Mg 2+ -ATPPase[(5.0±0.39)×10 -3 μmolPi/mgprot/hour]in epididymides of rats were higher significantly than that of Cd-induced group[(1.27±0.20)×10 -3 μmolPi/mgprot/hour、(0.27±0.52)×10 -3 μmoiPi/mgprot/hour and (3.55±0.15)×10 -3 μmolPi/mgprot/hour](P<0.05 or P<0.01).Conclusions Astragalus membranaceus injection can antagonize Cd-induced rats sperm motility toxicity.The effects might be related with protection of the activities of ATPase by Astragalus membranaceus.
