摘要
本试验旨在探讨sophyβ-1,3/1,6-葡聚糖对肠炎沙门氏菌感染Caco-2细胞抗氧化功能的影响。选用Caco-2细胞作为试验模型,分别用MEM(A组)、β-1,3/1,6-葡聚糖(B组)、肠炎沙门氏菌(C组)处理Caco-2细胞,以及先用β-1,3/1,6-葡聚糖预处理,再用肠炎沙门氏菌处理(D组),每个处理3个重复。结果表明,沙门氏菌感染3与12h时Caco-2细胞上清中总抗氧化酶(T-AOC)活性显著升高(P<0.05),与对照组相比,C组Caco-2细胞上清中丙二醛(MDA)水平在沙门氏菌感染3h时显著下降,但感染12h后显著升高(P<0.01);添加β-1,3/1,6-葡聚糖可以显著升高12h细胞上清中总抗氧化酶(T-AOC)活性(P<0.01),并且有降低12h细胞培养上清中MDA水平和增加超氧化物歧化酶(SOD)活力,以及降低细胞裂解液中GSH水平的趋势。结果提示,β-1,3/1,6-葡聚糖可以通过增强T-AOC活力和下调MDA水平来提高细胞抗氧化能力,缓解由肠炎沙门氏菌引起的过氧化损伤。
In this study,the experiment was conducted to investigate the effects of beta-1,3/1,6-glucan on anti-oxidative functions of Caco-2cells infected by Salmonella enteritidis.In the experiment,Caco-2cell line was used as an in vitro model.Caco-2cells were treated by MEM(group A),beta-1,3/1,6-glucan(group B),Salmonella enteritidis(group C),respectively;Caco-2cells in group D were pre-incubated withβ-1,3/1,6-glucan and then infected with SE.The results showed that the T-AOC concentration significantly increased in group C at 3and 12 hin cell culture supernatant(P<0.05).Compared with the control group,the MDA concentration decreased after Caco-2cells treated 3h,while enhanced at 12h(P<0.01)in cell culture supernatant in group C.Pre-treated with beta-1,3/1,6-glucan increased significantly the total antioxidative capacity of T-AOC in cultured supernatant of Caco-2cells at 12h(P<0.01)and had the tendency to decrease the MDA concentration and increase the SOD activity in cultured supernatant.And the GSH concentration decreased in group D in cell lysate at 12 h.These results suggested that beta-1,3/1,6-glucan can enhance the anti-oxidative function of Caco-2cells challenged with SE by increasingthe T-AOC activity and cutting down the MDA concentration.
