摘要 目的:分析MDCK(犬肾细胞,Madin-Darby canine kidney cells,MDCK)单克隆细胞的生长特性,确定MDCK单克隆细胞库的可用性。方法:通过台盼蓝拒染测定细胞复苏活率,依据细胞生长的快慢、好坏和观察细胞形态判定细胞的生长速度、状态和形态.结果:MDCK单克隆细胞平均复苏活率为0.954±0.016,2~3 d长满的细胞占92.1%.57.9%的细胞生长状态良好,具有多角上皮样、梭形成纤维样和圆形岛状样三种形态.结论:MDCK单克隆细胞库中细胞复苏活率达到0.954±0.016,92.1%的单克隆细胞株在2~3 d长至单层致密,细胞状态良好.库内细胞有三种形态,可为病毒敏感细胞基质筛选提供丰富的资源。 Objective This study aimed to analyze the growth characteristics of monoclonal cells of MDCK(Madin-Darby canine kidney cells,MDCK)and to determine the availability of MDCK monoclonal cell bank.Methods The cell resuscitation rate was determined by trypan rejection method,the speed of growth of cells were detected according to the growth rate of the cells.And cell growth state were evaluated based on the quality of the cells and growth morphology classification of MDNK monoclonal cells.Results The average recovery rate of MDCK monoclonal cells was 0.954±0.016.The cells that were overgrown in 2-3 days accounted for 92.1%and 57.9%of the cells grew well.There were three cell forms,namely polyhedral epithelium,fusiform fibroids and circular islands.Conclusion The cell resuscitation rate of MDCK monoclonal cell bank is 0.954±0.016.The 92.1% monoclonal cell line was dense to monolayer in 2-3 days,and the cell state was very well.There were three forms in the cell bank,which can provide screening for virus sensitive cell matrix.
