广西猪细小病毒6型VP1基因序列进化分析

Phylogenetic analysis of the VP1 gene of porcine parvoviruses 6 from swine samples in Guangxi

为研究广西地区是否存在猪细小病毒6型(PPV6)毒株流行,分析PPV6毒株遗传进化和分子特征,采用PCR技术对广西地区采集的猪组织样品进行PPV6基因片段检测。结果表明,广西地区猪群存在PPV6,感染率为36%(27/75),同时也存在着PPV6与猪圆环病毒2型(PCV2)混合感染现象,感染率为32%;5株PPV6 VP1基因与中国参考株TJ株、韩国参考株KSU1-AZ-2014株、美国参考株U18-4株、波兰参考株P15-1株核苷酸序列同源性分别为99.3%～99.8%,99.3%～99.8%,98.9%～99.8%,99.3%～99.8%;VP1基因遗传进化分析表明,PPV6包含两个基因亚群,5株PPV6与中国参考株BJ、SC、JS、TJ株及波兰参考株处在G2亚群,而美国U18-4株则单独处于一个G1亚群。本研究为中国PPV6毒株流行病学及其病毒致病性研究提供研究基础。

In order to investigate the genetic evolutionary distribution and molecular characteristics of the PPV6 strain in Guangxi,PPV6 gene fragments were amplified by PCR and sequenced.The results showed that the infection rate of PPV6 in Guangxi pigs was 36%(27/75)and that there was PPV6 and PCV2 co-infection in the pigs at the rate of 32%.5 strains of PPV6 VP1 shared 99.3%~99.8%,99.3%~99.8%,98.9%-99.8%and 99.3%~99.8%identities with the Chinese reference TJ strain,the South Korean KSU1-AZ-2014 strain,the US U18-4 strain,and the Poland P15-1 strain,respectively.The genetic analysis of the VP1 gene showed that PPV6 contained two subgroups of genes.The 5 strains of PPV6 belonged to the same subpopulation of G1 with the Chinese strains of BJ,SC,JS and TJ and the Poland strains,while the USA strains were in another subpopulation of G1 alone.This study provided new data that supported research on the pathogenesis of the PPV6 strain and its virulence in China.
