摘要
为了建立检测利巴韦林残留的化学发光酶免疫方法,对酶标抗原与磁标抗体的最佳稀释倍数进行优化,建立标准曲线,同时对方法的检测限、准确度和精密度进行评价。结果显示:50%抑制浓度(IC50)为2.263μg/L,线性范围是0.409~12.527μg/L;在动物组织、兽药、饲料中的检测限为2.0μg/kg,样本添加回收率为83.6%~94.7%,批内、批间相对标准偏差均<10%;对实际样本进行检测,与液相色谱-质谱/质谱法的结果无显著性差异(P>0.05)。该方法灵敏度高、操作简便,可广泛用于动物组织、兽药、饲料中利巴韦林残留量的测定。
The aim of the study is to establish a method for detection of ribavirin residues by chemiluminescent enzyme immunoassay(CLEIA).The dilution multiples of the enzyme-labeled antigen and magnetic labeled antibody were optimized,a standard curve was established,and the detection limit,accuracy and precision of the method were evaluated.The results showed that the 50%inhibitory concentration(IC50)was 2.263μg/L,and the linearity range was 0.409-12.527μg/L.The detection limit in animal tissues,veterinary drugs and feed were 2.0μg/kg.The recoveries ranged from 83.6%to 94.7%,with the intra-assay and inter-assay coefficients of variation lower than 10%.There was no significant difference between CLEIA and liquid chromatographytandem mass spectrometry in the detection of the actual samples(P>0.05).In general,the above method was sensitive and simple,and might be widely used for detection of ribavirin residues in animal tissues,veterinary drugs and feed.
