摘要
菌株I06-02230是从云南西双版纳药用植物三尖杉根际土中分离得到的一株具有绿脓杆菌SecA蛋白抑制活性的稀有放线菌.对菌株发酵条件、活性组分理化性质和分离流程进行了初步研究.立瓶发酵时,190r/min往复摇床,28℃培养,发酵时间168h,发酵液抑制活性最大,此时发酵液的pH值为8.3.活性组分在弱酸及弱碱性条件下均比较稳定,为脂溶性成分.采用硅胶柱层析和HPLC等方法初步分离,获得具脂溶性活性组分的粗品2230A.
A rare actinomycete strain,I06-2230,was isolated from the rhizosphere soil of the medicinal plant Cephalotaxus fortunei Hook in Xishuangbanna of Yunnan province,China.The fermentation broth of I06-02230was found to inhibit against SecA protein of Pseudomonas aeruginosa.A preliminary study was made of its fermentation conditions,bioactive components and separation processes.In shake flask culture,the greatest inhibitory activity was obtained with the optimum fermentative conditions:medium at pH 8.3,28℃,190rpm of shaking and culture for 168hours.Its active component was a liposoluble constituent,which was fairly stable under subacid and alkalescent conditons.Silica gel column chromatography and HPLC were used to separate and purify the active component,and a crude liposoluble product 2230Awas obtained.
