摘要
对采自长江水域四川-重庆段大鰭鳠Mystus macropterus线粒控制区的克隆测序,并与长吻鮠Leiocassis longirostris、圆尾拟鲿Pseudobagrus tenuis和光泽黄颡Pelteobagrus nitidus mtDNA控制区序列进行比较分析,发现大鳍鳠线粒体控制区序列有腺嘌呤(A)、胸腺嘧啶(T)含量高,鸟嘌呤(G)含量低的特点.同时,大鳍鳠线粒体控制区包括3个主要识别区域:扩展终止相关序列、中央保守序列和保守序列.对哺乳动物,鸟类及其他脊椎动物线粒体控制区序列的比较分析,大鳍鳠中央保守序列区CSB-D,CSB-E,CSB-F与哺乳动物和其他鱼类的中央保守区相一致;大鰭鳠保守序列区的CSB-3比CSB-2的变异大些,与一些鱼类相比较,也有显著的差别.大鳍鳠在不同的种属间存在大致相同的保守区段,其中CSB-D和GTGGG-box结构较保守.该研究可以为将来进行大鰭鳠的种群分化研究和资源保护研究提供基础.
The mtDNA control region of Mystus m acropterus (Bleeker ) which included Sichuan and Chongqing in Yangtze River were get by cloning and sequencing ,and compared with the mtDNA sequences of Leiocassis longirostris ,Pseudobagrus tenuis and Pelteobagrus nitidus .In analogy ,the conserved region of these three species are identified as extended terminal associated sequences ,central conserved sequences block and conserved sequences block .Based on the comparison of mtDNA control sequences block of mam ‐mals ,birds and other vertebrates ,we found that central conserved sequences block of M ystus macropterus including CSB‐D ,CSB‐E ,CSB‐F ,consistent with mammals and other fish .The variation of conserved se‐quences block of CSB‐3 was than CSB‐2’s in Mystus macropterus ,and compared with some fish ,there are significant differences .The sequences of CSB‐D and GTGGG‐box are stringently conserved while a few site variations are found in other regions among species .The sequences data may provide a foundation for fu‐ture study on population differentiation and resource conservation of this species .
