黄芪注射液对大鼠脑出血灶周围细胞Mn-SOD和Cyt-c表达的影响 被引量:7

Influence of the Astragalus Injection on Perihematomal Mitochondrial Mn-SOD and Cyt-c Expression in Rats

摘要 目的探讨黄芪注射液对大鼠脑出血灶周围细胞中锰超氧化物歧化酶(manganese superoxide dismutase,Mn-SOD)和细胞色素C(Cytochrome C,Cyt-c)表达的影响。方法将健康SD大鼠随机分为正常组、假手术组、模型组和治疗组;大鼠自体尾动脉不凝血注入法建立脑出血模型;采用黄嘌呤氧化酶法检测出血灶周围脑组织中Mn-SOD活性;免疫组化法和Western blot检测脑组织中Mn-SOD、Cyt-c的表达变化。结果出血模型组脑组织中Mn-SOD活性和表达量较正常组和假手术组均明显降低(P<0.05),Cyt-c的表达明显增加(P<0.05);Mn-SOD活性和Cyt-c的表达呈负相关性(r=-0.935,P=0.004);黄芪注射液治疗组的Mn-SOD活性和表达量较模型组在3d、7d时有明显提高(P<0.05),Cyt-c表达在各时间点均存在明显降低(P<0.05)。结论黄芪注射液可能通过提高脑出血灶周围细胞中Mn-SOD的活性和表达,减少Cyt-c的释放,改善线粒体功能障碍,从而抑制脑出血灶周围细胞的凋亡。 Objective To observe the effect of astragalus injection on perihematomal mitochondrial manganese superoxide dismutase( manganese superoxide dismutase,Mn-SOD) and cytochrome C( Cytochrome C, Cyt-c) in rats. Methods SD rats were randomly divided into four groups,namely the normal group,sham operation group,model group and treatment group. Cerebral hemorrhage moded was established with rat tail artery autologous blood injection. Mn-SOD activity was detected by xanthine oxidase perihematomal brain manganese superoxide dismutase and brain tissue Mn-SOD,cytochrome C( Cyt-c) expression were detected byimmunohistochemistry and Western blot analysis. Results Mn-SOD activity and expression in were significantly higher than those in normal group and sham group( P < 0. 05). Cyt-c was significantly increased in the brain hemorrhage model group( P < 0. 05). Mn-SOD activity and Cyt-c expression was negatively correlated( r =-0. 935,P = 0. 004). Compared with the model group,Mn-SOD activity and expression in astragalus treatment group at 3,7d were significantly increased( P < 0. 05). Cyt-c expression at each time point were significantly lower in astragalus treatment group( P < 0. 05). Conclusion Astragalus may by increasing perihematomal mitochondria Mn-SOD activity and expression,reducing Cyt-c release and improvingmitochondrial dysfunction,thereby inhibiting perihematomal apoptosis.
