摘要
目的探讨失荅刺知丸对大鼠脑缺血再灌注后脑梗死体积及海马区神经生长因子(nerve gronth factor,NGF)表达的影响。方法 75只SD大鼠随机均分为假手术组、局灶性脑缺血再灌注对照组和失荅刺知丸灌胃组,其中失荅刺知丸灌胃组又根据生药煎煮的药液浓缩剂量分为低、中、高3个亚组。线栓法制成大鼠大脑中动脉再灌注模型,TTC染色法检测脑组织梗死体积改变,免疫组化染色法检测脑缺血再灌注后海马区细胞NGF的表达。结果 TTC染色后表明,失荅刺知丸组3个亚组脑组织梗死体积均小于局灶性脑缺血再灌注组(P<0.05),而失荅刺知丸组的3个亚组之间比较,随着失荅刺知丸剂量的增加脑梗死体积减小(P<0.05)。NGF免疫组化染色显示,假手术组大鼠海马区神经元胞体及突起内有黄色的NGF阳性颗粒排列整齐;局灶性脑缺血再灌注组大鼠海马区各区及齿状回NGF阳性细胞数目较假手术组减少(P<0.05);失荅刺知丸组3个亚组大鼠海马区及齿状回NGF阳性细胞数目均较假手术组减少(P<0.05),较局灶性脑缺血再灌注组增高(P<0.05),且失荅刺知丸组3个亚组的NGF阳性细胞数随着失荅刺知丸剂量的增加而增高(P<0.05)。结论失荅刺知丸能够减少大鼠的脑组织梗死体积,并能够增强大鼠脑缺血再灌注后海马区NGF的表达,从而起到神经保护作用。
Objective To investigate effects of Shi Da Ci Zhi Wan on NGF expression in the rat cerebral ischemia-reperfusion cerebral infarction volume and the hippocampus. Methods 75 SD rats were divided randomly into model,focal cerebral-ischemia reperfusion control group and Shi Da Ci Zhi Wan lavage group.Using thread-tie method,middle cerebral artery ischemia-reperfusion model was established. The brain infarction volume change was obserbed by the TTC staining method. NGF detection of focal cerebral ischemia-reperfusion in rat hippocampus expression of cells was determined by immunohistochemical staining. Results TTC staining showed,cerebral infarction volume in three subgroups of Shi Da Ci Zhi Wan drug group were smaller than that of focal cerebral ischemia reperfusion group( P < 0. 05). Model NGF immunohistochemical study showed,control the rat hippocampus neuron cell body and neat model NGF positive particles swelled with yellow,focal cerebral ischemia reperfusion group in the rat hippocampus and dentate gyrus districts model NGF positive cells number decreased significantly compared with control group( P < 0. 05); Shi Da Ci Zhi Wan group three subgroups in the rat hippocampus and dentate gyrus model NGF positive cells number were significantly reduced in the control group( P < 0. 05) and were higher than in focal cerebral ischemia reperfusion group,and the Shi Da Ci Zhi Wan group three subgroups of NGF plays positive cells number as Shi Da Ci Zhi Wan increased with the amount of pills. Conclusion Shi Da Ci Zhi Wan could reduce the rat cerebral infaction volume of brain tissue and enhance the hippocampus after ischemia-reperfusion rats model NGF expression which has a protective effect on ischemia reperfusion.
