大鼠脊髓损伤后Caspase-9表达和脊髓神经细胞凋亡相关性研究 被引量:2

摘要 目的研究Allen’s法致大鼠脊髓损伤后不同时间点神经细胞凋亡与Caspase-9表达的关系及其意义。方法将30只成年SD大鼠,随机分为3组,每组10只。假手术组:空白组:不损伤脊髓组织,仅行椎板减压术;手术组:损伤脊髓组织。对3组SD大鼠术前及术后6h、12h、24h行CBS评分后取其脊髓组织进行HE染色观察组织形态、TUNEL检测脊髓凋亡细胞,免疫组化检测Caspase-9表达水平。结果 3组大鼠术前CBS评分均为100分,术后手术组评分降低(P<0.05),HE染色结果显示:手术组不同时间点脊髓组织均明显充血水肿,而假手术组未见明显改变。TUNEL检测结果显示:假手术组不同时间点仅有少量脊髓神经细胞凋亡,而手术组在不同时间点脊髓神经细胞凋亡数较其他两组升高(P<0.05),其中12h时凋亡细胞最多。免疫组化检测结果显示:与假手术组比较,手术后不同时间点损伤脊髓组织中Caspase-9的表达均升高,(P<0.05),12h时达最高峰。手术组损伤脊髓组织12h和24h的Caspase-9表达高于6h手术组(P<0.05);24h和12h之间Caspase-9的表达量差异无统计学意义(P>0.05)。结论大鼠脊髓损伤后脊髓神经细胞的凋亡可能与Caspase-9的表达有关。 Objective To investigaterelationship between the expression of Caspase- 9 and apoptosis of spinal nerve after spinal cord injury in rats at different time points. Methods 30 adult SD rats were randomly divided into 3 groups: blank group,control group( does not damage the spinal cord,only using laminectomy),surgery group( damage the spinal cord). Using CBS score before processing and at 6h,12 h,24h time points after surgery,specimens of the spinal cord were taken and HE staining,TUNEL staining and immunohistochemical detection of the expression of Caspase- 9 were determined. Results All CBS score were 100 before surgery. Comparing with blank group and control group,the CBS score was significantly reduced in surgery group after surgery. HE staining result showed that,comparing with blank group and control group,congestion and edema was seriousness in surgery group at different time point after surgery,TUNEL staining result showed that,comparing with blank group and control group,neuronal apoptosis was seriousness in surgery group at different time point after surgery,especially at 12 h time point. Immunohistochemical results of the Caspase-9 expression was similar to TUNEL staining. Conclusion The expression of Caspase- 9 may be related to neuronal apoptosis in spinal cord injury.
