摘要 目的通过检测丝裂厚语化蛋白激酶(MAPK)的主要家族成员ERK1/2、JNK和P38MAPK的活化形式,即磷酸化的ERK1/2(p-ERK1/2)、磷酸化的JNK(p-JNK)和磷酸化的P38MAPK(p-P38MAPK)在人皮肤基底细胞癌、Bowen病及正常对照组中的表达水平,探讨ERK1/2、JNK和P38MAPK在基底细胞癌和Bowen病发生、发展中的作用。方法运用免疫组化技术分别检测p-ERK1/2、p-JNK和p-P38MAPK3种蛋白在基底细胞癌、Bowen病以及正常人皮肤对照组中的表达差异。结果 p-ERK1/2、p-JNK和p-P38MAPK蛋白在皮肤基底细胞癌和Bowen病皮损中的阳性表达均高于正常皮肤(P<0.05);且3种蛋白在基底细胞癌中的表达均明显高于Bowen病皮损中的表达(P<0.05)。结论 MAPK中的ERK1/2、JNK和P38MAPK的过度激活可能参与了皮肤基底细胞癌和Bowen病的发生发展。 Objective To explore the role of ERK1 /2,JNK and P38 MAPK activation in the development of Basal cell carcinoma( BCC) and Bowen's disease by detecting the level of the activated form of the main family members of MAPK: ERK1 /2,JNK and P38 MAPK,namely the phosphorylation of ERK1 /2( p- ERK1 /2),phosphorylation of JNK( p- JNK) and phosphorylated P38MAPK( p- P38MAPK) in human Basal cell carcinoma,Bowen's disease and normal control group. Methods The expression of p- ERK1 /2,p- JNK and p- P38 MAPK proteins in BCC,Bowen's disease and normal skin were all detected by immunohistochemistry( IHC). Results Compared with human normal skin,the proteins of p- ERK1 /2,p- JNK and p-P38 MAPK were all over- expressed in BCC and Bowen's disease( P < 0. 05). The expression of the three proteins in BCC were significantly higher than that in Bowen's disease( P < 0. 05). Conclusion The abnormal activation of ERK1 /2,JNK and P38 MAPK may take part in the development of BCC and Bowen's disease.
