摘要
目的腹腔注射链脲佐菌素(streptozotocin,STZ)建立高血糖模型和双侧颈总动脉结扎加低血压(2VO)的方法建立的脑缺血模型,探究高血糖对脑缺血卒中后癫痫发作的影响。方法 SD雄性大鼠40只,分为假手术组、空白对照组、高血糖组、脑缺血组、高血糖脑缺血组。腹腔注射STZ建立高血糖模型,4周后在高血糖模型基础上用2VO的方法建立脑缺血模型,利用脑电图监测大鼠脑电的变化和原位末端转移酶标记技术(terminal deoxynucleotidyl transferase-mediated d UTP-biotin nick end labeling,TUNEL)检测大脑神经元凋亡状况。结果脑电图观察结果显示高血糖脑缺血组大鼠出现明显的高波幅棘波、尖波发放。TUNEL染色结果显示高血糖组、脑缺血组高血糖脑缺血组均出现大量凋亡神经元,并且高血糖脑缺血组凋亡的神经元数量明显高于其他组。结论高血糖能够加重脑缺血卒中后大鼠的癫痫发作和大脑神经元凋亡。
Objective To explore the influence of hyperglycemia on the post- stroke epilepsy rat model after cerebral ischemia. Methods 40 SD male rats were divide into 4 groups: control,sham operation,hyperglycemia,cerebral ischemia,cerebral ischemia with hyperglycemia. The hyperglycemia model was established by peritoneal injecting STZ,4 weeks later completing the cerebral ischemia model by ligaturing two- sides CCA and keeping low circulating volume( 2VO) of the rats. The rats’ electrical activity was detected by EEG and the by TUNEL. Results EEG showed that the cerebral ischemia with hyperglycemia group appeared the high amplitude spikes and sharp waves apparently. TUNEL showed that both the hyperglycemia group and the cerebral ischemia group appeared a large number of apoptotic neurons,and the amounts of the cerebral ischemia with hyperglycemia group were significantly higher than those of the other groups. Conclusion Hyperglycemia can aggravate the attack rate and cerebral injury.
