高效液相色谱法测定人血浆中依托泊苷浓度

摘要 目的建立人血浆中依托泊苷药物浓度的HPLC测定方法。方法采用高效液相色谱法。ZORBAX XDB-C18色谱柱(250mm×4.6mm,5.0μm);ZORBAX XDB-C18(4mm×2.0mm,5μm)保护柱,柱温30℃,流动相为乙腈-甲醇-水=5∶49.4∶45.6,流速为1.0m L·min-1,检测波长为230nm。结果在0.5~50μg·m L-1内线性方程为y=1.7563x-0.0134(r=0.9999,n=7)。低(1.0μg·m L-1)、中(5.0μg·m L-1)、高(40.0μg·m L-1)3个浓度的质控样本的批内变异(RSD)为0.4%~2.9%,批间变异(RSD)为2.4%~6.9%,方法学回收率为92.6%~106.6%。结论本方法灵敏度高、简便快捷,为开展依托泊苷的治疗药物浓度监测提供了有效的技术保障。
