糖原合酶激酶-3β的表达量变化对少突胶质前体细胞分化的影响

摘要 目的探讨糖原合酶激酶-3β(GSK3β)表达量的变化对少突胶质前体细胞(OPCs)分化的影响,以证明GSK3β参与了OPCs的正常分化过程。方法体外shRNA干扰OPCs的GSK3β基因表达,通过Western blot方法观察OPCs的分化情况。同时利用GSK3β的活化质粒,进行体外细胞转染实验,通过Western blot方法观察过表达GSK3β活化质粒对OPCs分化的影响。结果 Western blot实验结果表明,体外干扰GSK3β的表达后,成熟少突胶质细胞的标记物CNP蛋白表达量下降(P<0.05),表明OPCs的正常分化过程受到抑制,不能分化为成熟的少突胶质细胞。体外过表达GSK3β的活性形式时,细胞内非活性形式的p GSK3β表达量下降,CNP的表达量升高(P<0.05)。结论 GSK3β参与了少突胶质前体细胞的分化过程,具有促进OPCs分化的作用。 Objective To investigate the effect of GSK3β expression level changes on the development of oligodendrocyte precursor cells( OPCs) into the oligodendrocytes( OLs). Methods In vitro RNAi was used to interfere GSK3β gene expression and the effect on OPCs differentiation was observed by western blot. In vitro GSK3βactive plasmid was transfected into OPCs and the effect of over- expression GSK3β on OPCs development was observed by western blot. Results The western blot results showed that when GSK3β activity was inhibited,CNP expression level significantly decreased,indicating that the normal process of OPCs differentiation into OLs was blocked. However,when GSK3β was over- expressed,CNP expression level remarkably enhanced. Conclusion GSK3β takes part in the process of OPCs differentiation into OLs and it promotes this process.
