摘要
目的观察野生型小鼠和脂联素基因敲除(APN^(-/-))小鼠胰岛素抵抗状态对心肌细胞凋亡率的影响及脂联素干预作用。方法选择雄性野生小鼠16只和雄性APN^(-/-)小鼠24只,分5组,每组8只,分别为对照组(con)、胰岛素抵抗组(IR)、基因敲除组(KO)、基因敲除+胰岛素抵抗组(KIR)、基因敲除+胰岛素抵抗+脂联素干预组(APN)。高脂饲料诱导小鼠胰岛素抵抗模型。测空腹血糖(FPG)和空腹胰岛素(FINS)水平,计算胰岛素抵抗指数(HOME-IR),采用FITC-PI凋亡试剂盒测心肌细胞凋亡率。结果 (1)与con组比较,IR、KO组小鼠FINS、HOME-IR值以及心肌细胞凋亡率均升高(P均<0.05)。(2)与KO组相比,KIR组体重、FINS、HOME-IR值以及心肌细胞凋亡率均明显升高(P均<0.05)。(3)与KIR组相比,APN组小鼠FPG、FINS、HOME值及心肌细胞凋亡率降低(P<0.05)。结论胰岛素抵抗小鼠心肌细胞凋亡率升高,脂联素干预可以改善胰岛素抵抗,降低心肌细胞凋亡率。
Objective To observe effect of the insulin resistance and adiponectin intervention on myocardial cell apoptosis rate in wild type mice and adiponectin gene knockout( APN^(-/-)) mice. Methods Sixteen male wild mice and twenty- four APN^(-/-)mice were randomly divided into 5 groups,8 animals for each group: control group( con),insulin resistance group( IR),knockout group( KO),gene knock out and Insulin resistance group( KIR),adiponectin intervention group( APN). High fat feed induced insulin resistance model in mice. The level of fasting plasma glucose( FPG) and fasting insulin( FINS) were tested at the end of the experiment. The myocardial cell apoptosis rate were detected by FITC- PI apoptosis kit. Results( 1) Compared with control group,FPG,FINS,HOME- IR levels and myocardial apoptosis rate were significantly increased in the IR group and KIR group( P < 0. 05).( 2) Compared with KIR group,FPG,FINS,HOME- IR levels and myocardial apoptosis rate were significantly decreased in the APN group( P < 0. 05). Conclusion The cardiomyocytes apoptosis rate increased in mice with Insulin resistance. Adiponectin intervention can improve insulin resistance and decrease the rate of myocardial cell apoptosis.
