宁夏回汉族HSP70家族基因多态性与原发性高血压的关联研究

Association of HSP70 Gene Polymorphisms with Essential Hypertension in Hui and Han Groups of Ningxia

目的探讨热休克蛋白70(heat shock protein 70,HSP70)基因多态性与宁夏回汉民族人群原发性高血压(essential hypertension,EH)之间的关联。方法采用病例对照研究设计,按照纳入和排除标准筛查出270例无血缘关系的原发性高血压患者作为病例组,471例无血缘关系的健康人作为对照组。结合聚合酶链反应-限制性片段长度多态性(PCR-RFLP)技术确定研究对象HSP70-1基因(+190G/C)、HSP70-2基因(+1267A/G)和HSP70-hom基因(+2437T/C)的基因型,分析研究指标与EH患病的关系。结果 HSP70-hom基因的+2437位点基因型及等位基因频率分布在病例组与对照组之间差异均有统计学意义(χ~2=7.501、4.099,P均<0.05),病例组中T/C基因型及C等位基因的频率高于对照组。HSP70-2基因1267位点各基因型及等位基因频率分布在病例组回族与汉族间的差异均有统计学意义(χ~2=9.605、5.651,P<0.01或<0.05);不同腰臀比(WHR)水平下HSP70-hom基因+2437T/C等位基因频率在汉族中分布差异有统计学意义(χ~2=4.545,P<0.05)。结论 HSP70-hom基因+2437位点多态性与宁夏回汉民族原发性高血压有关,T/C基因型及C等位基因增加了携带者原发性高血压的发病风险;HSP70-2基因+1267位点A/A基因型及A等位基因增加了回族携带者原发性高血压的发病风险;G/G基因型及G等位基因增加了汉族携带者原发性高血压的发病风险。HSP70-hom基因+2437位点C等位基因增加了汉族携带者腹型肥胖发病的风险。

Objective To investigate the association between the polymorphisms of heat shock protein( HSP)70 family gene and essential hypertension( EH) in Hui and Han people of Ningxia. Methods A case- control study was conducted with 270 EH cases and 471 health controls. Polymerase chain reaction- restriction fragment length polymorphism technique( PCR- RFLP) was applied to detect the single nucleotide polymorphisms of HSP70- 1( + 190 G/C),HSP70- 2( + 1267 A/G) and HSP70- hom( + 2437 T/C). The association between the single nucleotide polymorphisms of HSP70 and EH was analyzed by Chi- square test,analysis of variance and logistic regression. Results There was significant difference in the distribution of genotypes and allele frequencies of HSP70- hom between the two groups( χ~2= 7. 501,4. 099,P all < 0. 05) and T/C genotype and C allele frequency in case group were significantly higher than those of controls. There was significant differences in the distribution of genotypes and allele frequencies of HSP70- 2( + 1267) in cases between Hui and Han ethnic group( χ~2= 9. 605,5. 651,P < 0. 01 or < 0. 05). In different level of WHR,the genes of HSP70- hom allele frequency showed significantly different distribution in the Han ethnicities( P < 0. 05). Conclusion There was significant correlation among HSP70- hom + 2437 with Essential hypertension in Ningxia. T/C genotype and C allele carriers increase the risk of essential hypertension in people.HSP70- 2 gene 1267- point A/A genotype and A allele carriers increase the risk of essential hypertension in Hui people while HSP70- 2 gene 1267- point G/G genotype and G allele carriers increase the risk of essential hypertension in Han people. HSP70- hom gene 2437 points C allele carriers increase risk of abdominal obesity between Hui and Han ethnicities.
