环磷酰胺致大鼠膀胱黏膜损伤及E-cadherin表达下调 被引量:2

摘要 目的探讨E-cadherin在环磷酰胺处理后膀胱黏膜中的作用。方法 SD大鼠12只,随机分为处理组和对照组,处理组大鼠给与环磷酰胺100mg·kg^(-1)腹腔注射,对照组给与同等剂量的生理盐水腹腔注射,24h后取膀胱,观察形态变化,检测E-cadherin mRNA表达水平、表达量。结果肉眼观处理组膀胱出现明显的水肿、出血、粘连;HE染色可见,膀胱黏膜弯曲减少、肿胀,上皮细胞层次减少,上皮变薄,膀胱黏膜呈现毛刺样改变;免疫组化可见,E-cadherin只表达在膀胱黏膜,处理组表达低于对照组;Real-time PCR与Western blot结果可见,E-cadherin mRNA与蛋白的表达量处理组均低于对照组(P<0.05)。结论大鼠腹腔注射环磷酰胺致上皮细胞严重损伤,E-cadherin在膀胱黏膜中表达下调。 Objective To explore the role of the E- cadherin in the bladder mucosa after cyclophosphamide treatment. Methods A total of 12 SD rats were randomly divided into two groups: treatment group( n = 6)and control group( n = 6). Rats in the treatment received intraperitoneal injection of cyclophosphamide 100 mg·kg^(-1)while rats in the control group received the same dose of intraperitoneal injection of saline. The bladders were collected after 24 hours to observe morphological changes and the change of morphological structure and E- cadherin protein expression,E- cadherin mRNA expression level. Results Bladder tissure of the treatment group showed visible edema,hemorrhage,adhesion; HE staining showed less bladder mucosa bending,swelling,arranged loosely cells and burr change. Expression of E- cadherin protein located in bladder mucosa by immunohistochemistry. Expression of E- cadherin mRNA in treatment group was lower than that of the control group by Real- time PCR( P < 0. 05). E- cadherin expression quantity in treatment group was significantly lower than that of the control group( P < 0. 05). Conclusion Rat peritoneal injection cyclophosphamide can induce E- cadherin expression down- regulation in bladder mucosa.
