摘要 目的探讨DC-CIK(dendritic cell-cytokine induced killer cell)过继细胞免疫治疗对原发性肝癌患者细胞免疫功能的影响。方法选取42例原发性肝癌患者,利用流式细胞术检测患者治疗前、后(5个治疗周期)外周血T淋巴细胞亚群及CD4^+CD25^+调节性T细胞;应用酶联免疫吸附试验检测血清IL-2、TNF-α、IFN-γ。结果经5个周期DC-CIK细胞免疫治疗,患者外周血T淋巴细胞亚群(CD3^+)、辅助性T细胞亚群(CD3^+CD4^+)、杀伤性T细胞亚群(CD3^+CD8^+)、NKT细胞亚群(CD3^+CD16^+56^+)的百分数与治疗前相比差异均无统计学意义(P>0.05);CD4^+CD25^+调节性T细胞百分数较治疗前降低(P<0.05)。患者血清IL-2、TNF-α、IFN-γ水平较治疗前提高(P<0.05)。结论 DC-CIK细胞免疫治疗可降低原发性肝癌患者外周血CD4^+CD25^+调节性T细胞的百分数,提高患者Th1类细胞因子水平,是原发性肝癌患者重要的辅助治疗手段。 Objective To explore the effects of DC- CIK on cellular immune function in patients with primary liver cancer,who accepted DC- CIK( dendritic cell- cytokine induced killer cell) adoptive cellular immunotherapy. Methods From 2012 March to 2014 December,42 cases with primary liver cancer in tumor biotherapy center of Ningxia people's Hospital for treatment were recruited in this study,who accepted DC- CIK treatment( 5 cycles). Flow cytometry was applied to detect peripheral blood T lymphocyte subsets and CD4^+CD25^+and ELISA was used to detect IL- 2,TNF- α,IFN- γ. Results After 5 cycles of DC- CIK cell immunity treatment,no significant difference of peripheral blood T lymphocyte subsets( CD3^+),helper T cell subsets( CD3^+CD4^+),cytotoxic T cell subsets( CD3^+CD8^+),NKT cell subsets( CD3^+CD16^+56^+) in patients were found between before and after treatment( P > 0. 05). CD4^+CD25^+regulatory T cell levels were significantly decreased than those before treatment( P < 0. 05). The IL- 2,TNF- α,IFN- γ levels of patients were significantly improved than those before treatment( P < 0. 05). Conclusion DC- CIK cell immunotherapy can decrease peripheral blood CD4^+CD25^+regulatory T cell percentage and improve Th1 cytokines levels in patients with the primary hepatocellular carcinoma and it is an important auxiliary treatment in patients with primary liver cancer.
