羟丙基-β-环糊精对白藜芦醇的包合作用及其包合物对小鼠游泳耐力的影响

摘要 目的研究羟丙基-β-环糊精对白藜芦醇的包合作用,以及白藜芦醇-羟丙基-β-环糊精包合物对小鼠运动耐力的影响。方法 1)采用紫外光谱法和核磁共振法,探讨羟丙基-β-环糊精对白藜芦醇的包合作用;另外,采用相溶解度法,计算羟丙基-β-环糊精对白藜芦醇的增溶倍数。2)雄性昆明小鼠随机分为:A组(安静对照组)、B组(训练组)、C组(白藜芦醇训练组)、D组(白藜芦醇-羟丙基-β-环糊精包合物训练组),在6周的递进式游泳训练后,进行小鼠力竭游泳实验。结果 1)羟丙基-β-环糊精对白藜芦醇有包合作用,能明显提高白藜芦醇的水溶解度(S=0.0140 g·L^(-1),S'=0.5900 g·L^(-1)),提高了近40倍;2)小鼠力竭游泳时间B、C、D组长于A组,C、D组长于B组,D组长于C组(P均<0.01)。结论 1)白藜芦醇能与羟丙基-β-环糊精形成包合物,显著提高其溶解度;2)白藜芦醇-羟丙基-β-环糊精包合物提高小鼠游泳运动耐力的作用优于白藜芦醇单体。 Objective To explore the inclusion complexation of resveratrol with hydroxypropyl- β- cyclodextrin,and to investigate effect of their complexes on exercise capacity of mouse. Methods 1) The inclusion complexation of resveratrol with hydroxypropyl- β- cyclodextrin was investigated by UV- vis spectrophotometer and NMR while the solubilization of the product was calculated by phase- solubility method. 2) Male Kunming mouse were selected and divided randomly into: group A( control group),group B( endurance exercise group),group C( endurance exercise group of resveratrol) and group D( endurance exercise group of complexes). After swimming training for six weeks,the duration of exhaustive swimming was investigated.Results 1) The inclusion complexation of resveratrol with hydroxypropyl- β- cyclodextrin was found,and the phase- solubility studies indicated that the solubility of resveratrol was substantially increased 40- folds in water. 2) The time of exhaustive swimming of group D was significantly longer than that of group C( P <0. 01). Conclusion 1) The B benzene ring of resveratrol penetrated into the wide cavity of hydroxypropyl-β- cyclodextrin,and resulted in the formation of inclusion complexes. 2) The complexes of resveratrol can lengthen the duration of exhaustive swimming longer than resveratrol can,and it improve greatly the exercise capacity of mouse.
