摘要
目的 通过观察羊颞下颌关节(temporomandibular joint,TMJ)间接创伤后不同时期髁突软骨中基质金属蛋白酶-13(matrix metalloproteinase-13,MMP-13)的表达,探讨MMP-13在TMJ创伤后导致颞下颌关节骨关节病(temporomandibular joint osteoarthrosis,TMJOA)过程中的作用。方法 用自制撞击装置造成山羊双侧颞颌关节间接性创伤。分别于伤后1周、1个月、3个月、6个月取材,并以正常TMJ作为对照,用免疫荧光组织化学方法进行观察。结果 TMJ髁突软骨中MMP-13伤后1周即出现强的表达,以后逐渐减弱;荧光着色于软骨破坏区增生滑膜样组织及软骨肥大带较强,正常对照组基本呈阴性反应。结论 MMP-13参与了TMJ创伤所致TMJOA过程,并起了重要作用。
Objective In order to understand the roles of matrix metalloproteinase - 13 ( MMP - 13) in the pathogen-esis of temporomandibular joint (TMJ) after indirect trauma. Methods Trauma to the TMJ in 25 goats was achieved under an impact to the right and left mandibular angle with a self-made device. The goats were killed 1w, 1 month, 3 months and 6 months after impact respectively. The expression of MMP - 13 in the condyle of TMJ was observed by immunofluores-cence histochemical staining. The normal TMJs were used as controls. Results The strong positive expression of MMP -13 was observed in the condlye of TMJ early after indirect trauma. The stronger expression was observed in the hypertrophic layer and the synovium-like tissue of destroyed area of condyle. Conclusion MMP - 13 may play an important role in the pathogenesis of TMJ after indirect trauma.
