摘要
【目的】观察挤压伤大鼠血清对培养的心脏成纤维细胞的增殖作用。【方法】培养1~3d龄SD大鼠心脏成纤维细胞,观察挤压伤大鼠血清和正常大鼠血清对培养的心脏成纤维细胞数量、蛋白质含量、犤3H犦-胸腺嘧啶核苷(犤3H犦-TdR)和犤3H犦-脯氨酸(犤3H犦-Proline)掺入的影响。【结果】与正常大鼠血清组比较,挤压伤大鼠血清使培养的心脏成纤维细胞的数量(×109/L)、总蛋白含量(μg/2mL)分别由0.33±0.01、163.3±24.1增加至0.50±0.02和324.6±52.2,犤3H犦-Proline和犤3H犦-TdR掺入后测得的每分钟脉冲数则分别由1211.7±140.7、1104.8±113.9上升至2000.7±96.2和2066.2±223.5。【结论】肢体挤压伤大鼠血清可促进培养的心脏成纤维细胞增殖和胶原合成。
To investigate the effects of crush injury rat serum on cell proliferation in cultured neonatal rat cardiac fibroblasts. 1 3 d old neonatal rat cardiac fibroblasts were cultured in vitro and the effects of crush injury rat serum on cell numbers, total protein content,-TdR and -Proline incorporation were observed in cultured rat cardiac fibroblasts. Compared with normal rat serum group,crush injury rat serum increased cell numbers(×109 /L) and total protein contents (μg/2mL) of cardiac fibroblasts from 0.33±0.01,163.3±24.1 to 0.50±0.02 and 324.6±52.2,the CPM of -Proline and -TdR after incorporation increased from 1 211.7±140.7,1 104.8±113.9 to 2 000.7±96.2, 2 066.2±223.5.[Conclusion]Crush injury rat serum can increase cell proliferation and collagen synthesis in cultured rat cardiac fibroblasts.
