大鼠脑缺血预处理后脑组织胱天蛋白酶-3蛋白的表达

摘要 目的 探讨胱天蛋白酶 3(caspase 3)在脑缺血预处理过程中的表达和意义。方法 用免疫组织化学染色技术分别检测假手术对照组、缺血预处理对照组、缺血组和缺血预处理组大鼠脑组织石蜡切片中caspase 3蛋白的表达情况。结果 缺血组和缺血预处理组CA1区caspase 3蛋白免疫反应强度明显高于假手术对照组 ;缺血预处理组再灌注 4 8和 72h与再灌注时间相同的缺血组比较 ,caspase 3蛋白免疫反应强度差异有显著性意义。结论 脑缺血预处理的保护机制可能与抑制caspase 3蛋白表达、减少脑细胞凋亡有关。 Objective To study the expression of caspase-3 protein in rat brain subjected to cerebral ischemic preconditioning.Methods Immunohistochemical technique was used to detect caspase-3 expression in rat brain sections obtained from the following groups:the sham operation group,the preconditioning group,the cerebral ischemic group and the cerebral preconditioning-ischemic group.Results The caspase-3 expression in both cerebral ischemic group and cerebral preconditioning-ischemic group was significantly higher than that in sham operation group (P<0.01).The caspase-3 expression in cerebral ischemic group after reperfusion for 48 h and 72 h was significantly higher than that in cerebral preconditioning-ischemic group after reperfusion for 48 h and 72 h,respectively(P<0.05,P<0.01).Conclusions The protective mechanism of ischemic preconditioning may contribute to inhibiting caspase-3 expression and subsequently inhibiting cerebral cell apoptosis.
