胶束薄层扫描法测定黄连及其制剂中的小檗碱、巴马汀和药根碱 被引量:17

Determination of Berberine, Palmatine and Jatrorrhizine in Coptis and Its Preparations by Micellar Thin Layer Chromatographic Scanning Method

摘要 以 2 %CPC 醋酸乙酯 (9∶1)胶束溶液为展开剂 ,在聚酰胺薄膜上成功地分离了小檗碱、巴马汀和药根碱。以 345nm为测定波长 ,5 5 0nm为参比波长进行扫描测定 ,建立了一种新的同时测定黄连及其制剂中小檗碱、巴马汀和药根碱的胶束薄层扫描法。小檗碱、巴马汀和药根碱的线性范围分别为 0 .2~ 2 .4、0 .1~ 1 0和0 .1~ 1.0 μg ;回收率分别为 97.7%~ 99.4 %、10 1.7%~ 10 2 .6 %和 96 .7%~ 97.8% ;相对标准偏差分别为 1.4 %~ 1.7%、1.9%~ 2 .7%和 1.8%~ 2 .2 %。 A new micellar thin layer chromatographic scanning method has developed for the simultaneous determination of berberine, palmatine and jatrorrhizine in coptis and its preparations. Using a mobile phase of 2% cetylpyridinium chloride (CPC)-ethyl acetate (9∶1) micellar solution and using polyamide film thin-layer, the berberine, palmatine and jatrorrhizine were well separated from each other and the R f values of them were 0.46, 0.69 and 0.35 respectively. The spots were scanned at 345 nm with reference wavelength of 550nm. The linear ranges of calibration curve were 0.2~2.4 μg/spot for berberine, 0.1~1.0μg/spot for palmatine and jatrorrhizine. The recovery of berberine, palmatine and jatrorrhizine were 97.7%~99.4%, 101.7%~102.6% and 96.7%~97.8% respectively, and their relative standard deviations were 1.4%~1 7%, 1.9%~2.7% and 1.8%~2.2% respectively.
