摘要
以壳寡糖与对羟基苯甲醛反应合成的席夫碱化合物作为配体,构建了一系列的金属Cu(Ⅱ)配合物,并通过紫外光谱法、电化学手段以及粘度实验,初步研究了配合物与DNA的作用机理。紫外光谱实验发现,配合物的特征吸收峰在加入DNA后峰位均呈现红移和减色;从循环伏安曲线可见,DNA加入后配合物的氧化峰电流呈下降趋势,峰电位伴随有正移趋势;DNA的粘度随着配合物浓度的依次增大,呈先增大后降低的趋势。结合以上这些信息,认为配合物与DNA以部分嵌插方式作用的。
A new Schiff base with good biological activity was synthesized by the reaction of oligo chitosan with p-hydroxybenzadehyde, and acted it as ligand to prepare Schiff base-copper complexes. Moreover, the interaction of Schiff base copper complex with herring sperm DNA was studied by cyclic voltammetry, UV-Vis spectra and viscosity methods. UV-Vis spectra result showed that the ultraviolet absorption intensity of complex was decreased by DNA, and adding DNA into the Schiff base copper complex solution, the redox peak current decreased and the peak potential shifted to positive. The viscosity raise at lower concentration and decreased at higher concentration. All results showed that the Schiff base copper complex and DNA was interacted partly according to the mode of intercalation.
