类叶升麻苷对脑缺血再灌注大鼠学习记忆能力及炎症因子的影响 被引量:4

Effects of Acteoside on Learning and Memory Ability and Inflammatory Factors in Rats with Cerebral Ischemia Reperfusion

摘要 目的观察类叶升麻苷对脑缺血再灌注大鼠学习记忆及炎症因子的影响,探讨其相关作用机制。方法 30只SD雄性大鼠随机分为假手术组、模型组、尼莫地平组和类叶升麻苷低、高剂量组,每组6只。采用线栓法制作大脑中动脉栓塞慢性缺血再灌注痴呆大鼠模型,通过跳台、Y迷宫实验检测大鼠学习记忆能力,采用流式细胞仪及ELISA试剂盒测定血清、海马及皮层中白细胞介素(IL)-1α、IL-2、肿瘤坏死因子(TNF)、干扰素-γ(IFN-γ)的含量。结果 Y迷宫实验结果显示:与假手术组比较,模型组大鼠新异臂进入次数减少,其他臂进入次数增加,新异臂潜伏期增加(P<0.05,P<0.01);与模型组比较,尼莫地平组和类叶升麻苷低、高剂量组大鼠新异臂进入次数增加,其他臂进入次数减少,新异臂潜伏期减少(P<0.05,P<0.01)。跳台实验结果显示:与假手术组比较,模型组大鼠测试期潜伏期减少,犯错误次数增加(P<0.01);与模型组比较,类叶升麻苷低、高剂量组大鼠测试期潜伏期增加,犯错误次数减少(P<0.05,P<0.01);流式细胞仪及ELISA试剂盒检测显示:与假手术组比较,模型组大鼠血清、海马和皮层IL-1α、IL-2、TNF含量明显增加(P<0.05,P<0.01),海马IFN-γ含量明显减少(P<0.05);与模型组比较,类叶升麻苷低、高剂量组大鼠血清、海马和皮层IL-1α、TNF含量明显降低,血清、海马IL-2明显降低(P<0.05,P<0.01),海马IFN-γ含量明显增加(P<0.01)。结论类叶升麻苷可提高MCAO大鼠学习记忆能力,且可有效降低模型大鼠炎症因子水平,其对模型大鼠的防治作用可能与抑制炎症反应有关。 Objective To study the effects of acteoside on learning and memory ability and IFNlammatory factors in rats with cerebral ischemia reperfusion;To discuss its mechanism of action. Methods Totally 30 SD rats were randomly divided into sham-operation group, model group, nimodipine group, acteoside low-and high-dosage groups, with 6 rats in each group. The rat model of dementia with chronic ischemia reperfusion of middle cerebral artery occlusion was established by using thread occlusion method. The learning and memory ability of rats were detected by means of step-down test and Y-maze test. The levels of IL-1α, IL-2, TNF and IFN-γ in serum, hippocampus and cortex were measured by flow cytometry and ELISA kit. Results The results of Y maze test showed that compared with the sham-operation group, the number of entry times of the new arms decreased, the times of other arms entered increased and the latency of the new arms increased in the model group(P<0.05, P<0.01). Compared with the model group, the number of entry times of the new arms increased, the times of other arms entered decreased and the loctency of the new arms decreased in nimodipine group, acteoside low-and high-dosage group(P<0.05, P<0.01). The results of step-down test showed that compared with the sham-operation group, the latency of the rats in the acteoside low-and high-dosage groups decreased and the number of mistakes decreased(P<0.01). Compared with the model group, the latency of the rats in the acteoside low-and high-dosage group increased and the number of mistakes increased(P<0.05, P<0.01). Flow cytometry and ELISA kit detection displayed that compared with sham-operation group, levels of IL-1α, IL-2, and TNF in serum, hippocampus and cortex in the model group significantly increased(P<0.05, P<0.01), and the content of IFN-γ in the hippocampus significantly decreased(P<0.05). Compared with the model group, serum, hippocampus and cortex levels of IL-1α in acteoside low-and high-dosage groups significantly decreased, and IL-2 in serum and hippocampus also significantly decreased(P<0.05, P<0.01), the content of IFN-γ in hippocampus significantly increased(P<0.01). Conclusion Acteoside can improve the learning and memory ability of MCAO cerebral ischemia reperfusion rats, and can effectively reduce the level of inflammatory factors. The preventive and therapeutic effects on vascular dementia rats may be related to the inhibition of inflammatory response.
