摘要
目的研究全蝎白术混合发酵品的抗肿瘤活性,确定药物抗肿瘤的活性部位。方法依据双指标优化提取工艺的最优条件对全蝎白术混合发酵品进行超声提取。采用超滤手段,分别用截留量为5kD和10k D的超滤膜对水提液进行超滤分段,测定各分子量段中蛋白质和多糖的含量,采用MTT法检测体外抗肿瘤活性,并以未发酵品做对照。结果各分子量段对2种肿瘤细胞抑制效果具有差异性,醇沉上清液对肿瘤细胞的抑制效果最好,其次为分子量段>10 kD全蝎白术混合发酵品,且全蝎白术混合发酵品对肿瘤细胞抑制作用显著优于未发酵品。结论全蝎白术混合发酵品对乳腺癌细胞、喉癌细胞均有较高的抑制作用,其活性部位主要为醇沉上清液和分子量段>10 kD全蝎白术混合发酵品。
Objective To study the antitumor activity of Scorpio and Atractylodis Macrocephalae Rhizoma mixed fermentation product;To determine the antitumor active sites of Scorpio and Atractylodis Macrocephalae Rhizoma mixed fermentation product.Methods Scorpio and Atractylodis Macrocephalae Rhizoma mixed fermentation product was extracted by ultrasonic assay on the basis of the optimal conditions of the double index optimization extraction process.The water extract was segmented by ultrafiltration membrane with a retention of 5 kD and 10 kD respectively.The contents of protein and polysaccharide in each molecular weight segment were determined.MTT assay was used to study the antitumor activity in vitro.The unfermented product was used as the control group.Results The inhibitory effects of different molecular weight segment on the two tumor cells were different,and the inhibitory effects of alcohol supernatant on the tumor cells were the best,followed by the SAHFP of molecular weight>10 kD segment.Scorpio and Atractylodis Macrocephalae Rhizoma mixed fermentation product was significantly better than that of unfermented products.Conclusion Scorpio and Atractylodis Macrocephalae Rhizoma mixed fermentation product has a high inhibitory effect on breast cancer cells and laryngeal cancer cells.The active sites of antitumor drugs are mainly the alcohol supernatant and the Scorpio and Atractylodis Macrocephalae Rhizoma mixed fermentation product of molecular weight>10 kD segment.
