摘要
为研究扶正化瘀方 (主药 :丹参、桃仁、人工虫草菌丝等 )治疗肝纤维化的机理 ,60只Wistar雄性大鼠 ,除 8只作为正常对照组 (A ,腹腔注射生理盐水 )外 ,5 2只大鼠腹腔注射 0 5 %对二甲基亚硝胺 (共 4周 ,每周 3次 ,每次 2ml/kg) ,造成肝纤维化模型 ,并随机分为 3组 ,模型对照组 (B ,18只 )、扶正化瘀方组 (C ,17只 )、秋水仙碱组 (D ,17只 ) ,分别给予自来水以及相应中西药物灌胃 ,共 4周。之后 ,处死大鼠取材检测各项指标。结果 :C组大鼠Hyp和肝组织胶原分期下降 ,纤维化程度减轻 ;活化HSC减少 ,凋亡增加 ;肝组织MMP 2、MMP 9、TIMP 2表达下降。结论 :促进活化HSC的凋亡、减少活化HSC的数量 ,并由此降低MMP 2、MMP 9、TIMP 2的表达、减轻肝纤维化程度 。
To explore the mechanism of 'Health-Supporting and Stasis-Resolving Decoction' in the treatment of liver fibrosis,52 male Wistar rats were given intra-abdominal injection of 0.5% p-dimethyl-nitroamine(2ml/2kg,three times a week for 4 weeks)to induce liver fibrosis,and then randomized into three groups:group B in which 18 rats were given tap water,group C in which 17 rats were given the Decoction and group D in which 17 rats were given colchicine;moreover,another 8 rats in group A were used as control.Four weeks later,all rats were sacrificed and relevant indexes were detected.Results:The Hyp and liver tissue collagen phase as well as fibrosis reduced;activated HSC decreased and apoptosis increased;the MMP-2,MMP-9 and TIMP-2 expressions reduced.Conclusion:The Decoction can promote the apoptosis of activated HSC and reduce activated HSC to reduce MMP-2,MMP-9 and TIMP-2 expressions,as a result to relieve liver fibrosis.
