HPLC与比色法测定复方黄芪注射液中总皂苷与黄芪甲苷含量

Determination of AstragalosidelV And Total Saponins In Fufang Huangqi Injection by HPLC and Colorimetry

目的:建立复方黄芪注射液中黄芪总皂苷与黄芪甲苷含量的测定方法。方法:黄芪总皂苷采用比色法(540nm,722型分光光度计);黄芪甲苷采用 HPLC—ELSD 法,Novapark^(?)C_(18)(3.9*150mm,4μm)色谱柱,流动相为乙腈—水(36:64),流速为1ml/min,柱温30℃,ELSD 参数:移管温度为105℃,气体流量1.50SLPM。结果:总皂苷平均回收率为100.64％,RSD 为1.74％;黄芪甲苷平均回收率为99.62％,RSD 为1.57％。结论:方法简便、快捷、准确、可靠,可用于复方黄芪注射液的质量控制。

Objective :To establish HPLC method and Colormetry for the determination of as- tragaloside Ⅳ and total saponin in Fufang Huangqi Injection.Methods.The total saponin of Radix Astragali adopted Colorimetry (540nm,722—Spectrophotometers );HPLC—ELSD method was used by astragaloside Ⅳ ,with a Novapark ^(?)C_(18)(3.9*150mm,4μm) ,acetonitrile—water(36:64)as mobile phase with a flow rate of 1.0ml/min,the temperature of Column and drift tube were 30℃, 90℃ respeceively,carrier gas flow 1.50SLPM.Results:The average recoveries of total saponin and astragaloside Ⅳ were 100.64%,99.62% respectively,with RSD of 1.74%,1.57%.Conclusion: It was proved that these methods were arrcurate and reliable for the quality control of Fufang Huangqi Injection.
