HPLC测定4种藏药中没食子酸的含量

Determination of gallinic acid in 4 Tibetan medicines by HPLC

目的 建立 4种藏药中没食子酸的含量测定方法。方法 采用HPLC法 ,ODS色谱柱 (2 0 0mm× 4 .6mm ,5 μm ,Dikma) ;流动相为甲醇 - 0 .1%磷酸 (5∶95 ) ;流速 1ml·min-1;波长 2 80nm。结果 没食子酸在浓度 0 .1～ 15 0 μg·ml-1范围内线性关系良好 (r≥ 0 .9991) ,平均回收率为 98%～ 10 1% ,精密度为 1%～ 3% ,重复性为 2 %～ 4 %。结论 所建立的没食子酸的HPLC法适用于 4种藏药的质量控制。

OBJECTIVE To establish a method for determination of gallinic acid contents in 4 Tibetan medicines.METHODS Gallinic acid was separated by HPLC.Adikma ODS column (200 mm×4.6 mm,5 μm) was used with mobile phase of methanol-0.1%phasphoric acid( 5∶95);The flow rate was 1 ml·min -1 and the detection wavelength was 280 nm.The sample powder was weighted and put into the 25 ml flask,in which the sample was dissolved with methanol,stirred and exfrated by ultrasound for 20 min.After fiftered by 0.45 μm micro-membrane,10 μl of the sample was injected in HPLC.RESULTS The linearity range was 0.1-150 μg·ml -1(r≥0.9991).The precision within day and between day and recovery were 1%-3%,2%-4% and 98%-101%,respectively.CONCLUSION The method could be applied to determine gallinic acid in 4 Tibetan medicines.
