Expression and clinical significance of matrix metalloproteinase-2 and CD147 in bladder transitional cell carcinoma

摘要 目的 通过检测基质金属蛋白酶-2(MMP-2)和CD147在膀胱移行细胞癌中的表达,探讨其在浸润转移中的作用。方法 用免疫组织化学链霉素抗生物素蛋白-过氧化酶法检测45例膀胱癌组织和9例正常膀胱粘膜中MMP-2及CD147的表达,并与临床病理指标进行对照分析。结果 膀胱癌组织中 MMP-2和 CD147过度表达,阳性率分别为 78%和 73%,MMP-2以浸润前缘的间质细胞表达为主,CD147表达于癌细胞。两者的联合表达率为66.7%,r=0.605(P<0.01)。结论 MMP-2和CD147与膀胱癌的恶性表型密切相关,间质细胞来源的MMP-2可能在膀胱癌的浸润转移中起主要作用。CD147与MMP-2的产生呈正相关,是膀胱癌潜在的治疗靶点。 Objective To investigate the expression of matrix metalloproteinase-2 and CD147 protein in bladder transitional cell carcinoma (BTCC) and its clinical significance. Methods The expres- sion of MMP-2 and CD147 protein was detected in 45 cases of bladder carcinoma and 9 cases of normal bladder mucosa by immunohistochemistrical method. The relationship between MMP-2 and CD147 ex- pression and clinicopathologic parameters were analyzed. Results MMP-2 and CD147 in specimens of bladder transitional cell carcinoma were over expressed with the positive staining rate being 78% and 73%, respectively. Immunohistostaining against MMP-2 was distributed prominently in stromal cells and much stronger in the invasive edge, but immunohistostaining against CD147 was distributed predominant- ly in cancer cells. Coexpression rate of MMP-2 and CD147 in bladder carcinoma was 66. 7% (r= 0. 605, P< 0. 01). Conclusion Overexpression of MMP-2 and CD147 was closely associated with malignant phenotype. MMP-2 derived of stromal cells may play a key role in invasion and metastasis of bladder car- cinoma. CD147 was significantly positively associated with expression of MMP-2, and it may be a promis- ing target molecule to prevent progression of bladder carcinoma.
