摘要
目的 研究MDM 2 (murinedoubleminute 2 )和 p5 3在卵巢肿瘤组织、细胞系表达及与卵巢癌发生关系。 方法 ①用细胞培养、蛋白质提取、Western免疫印迹法检测 17个卵巢癌细胞系MDM 2、p5 3蛋白表达。②用免疫组化SP法检测卵巢良性上皮性肿瘤、卵巢交界性上皮性肿瘤和卵巢癌中MDM 2、p5 3蛋白的表达。结果 ① 17个卵巢癌细胞系MDM 2蛋白p90表达阳性率为 11 8% (2 / 17) ;p5 3的阳性表达率为 3 5 3 % (6/ 17) ,2例MDM 2表达阳性出现在野生型卵巢癌细胞株。②在卵巢良性上皮性肿瘤、卵巢交界性上皮性肿瘤和卵巢癌中MDM 2的阳性表达率分别为 :2 0 % (2 / 10 )、2 5 % (2 / 8)和 2 5 % (5 / 2 0 ) ;p5 3的阳性表达率分别为 2 0 % (2 / 10 )、3 7 5 % (3 / 8)和 60 % (12 / 2 0 )。 3组中 ,MDM 2阳性表达率无显著性差异 (P >0 0 5 ) ,而 p5 3在卵巢癌中的阳性表达率显著高于卵巢良性上皮性肿瘤和交界性上皮性肿瘤 (P <0 0 5 )。结论 p5 3的异常表达与卵巢肿瘤的恶性表型相关。在卵巢癌的发生发展过程中 ,p5 3的异常表达比MDM 2更有意义。MDM 2表达阳性可能在无 p5 3基因突变的卵巢癌细胞发生。
Objective: To study the expression and relationship of MDM2 and p53 protein in ovarian tumor tissues? ovarian cancer cell lines. Methods: Cell culture?protein extraction?Western blotting and immunohistochemistry were used. Results: The rate of position expression of MDM2 protein in 17 ovarian cancer cell lines was 11 8%(2/17); the rate of positive expression of p53 was 35 3%(6/17). Two cases of positive expression of MDM2 only appeared in wild type p53, no position in mutation type. In ovarian benign epithelial tumors, borderline epithelial tumors and malignant epithelial tumors, the positive rates of MDN2 and p53 were 20%, 25%, 25%; and 20%, 37 5%, 60% respectively. The positive detection of MDM2 between the three groups of ovarian tumors was not significantly different (P>0 05), but the expression of p53 in malignant epithelial tumors was significantly higher than those in the other groups (P<0 05). Conclusion: The expression of p53 correlates with the malignant phenotypes of ovarian epithelial tumors. In the pathogenesis and development of malignant ovarian epithelial tumors, the abnormal expression of p53 may be more significant than that of MDM2. MDM2 might play a role in the ovarian cancer without p53 mutation.
