MMP-9和uPA在胃癌组织中的表达及相关性研究

Study on the Expression and Correlation of MMP-9 and uPA in Gas tric Cancer

目的 通过观察MMP 9和uPA在胃癌组织中的不同表达 ,分析它们在各种组织类型及不同分化程度的胃癌中的表达规律 ,探讨它们的异常表达与胃癌侵袭转移的关系。方法 采用免疫组织化学SP法分别检测MMP 9和uPA在 67例胃癌组织中的表达。结果 1) 67例胃癌组织中MMP 9阳性表达 3 4例 (50 .75% ) ,uPA阳性表达 3 5例 (52 .2 4% )。 2 )MMP 9、uPA在低分化胃癌中的表达率均比高分化中的高。 3 )有淋巴结转移者的MMP 9、uPA阳性率比无转移者显著增高。 4)早期胃癌组、侵及肌层组和侵犯全层组MMP 9阳性表达率分别为 7.69%、45.0 0 %、70 .59% ,差别有显著差异 (P <0 .0 1)。uPA亦然。 5)胃癌中MMP 9和uPA的表达呈正相关 (P <0 .0 1)。结论 MMP 9、uPA的表达均与胃癌的分化程度、浸润深度和淋巴结转移有关 。

Objective To study the expression of urokinase-type plasminogen activator (uPA) and matrix metalloproteinase-9 (MMP-9) on the inf iltration and metastasis of gastric cancer and to investigate the relationship o f their expression. Methods Immunohistochemical technique (SP m ethod) was applied to detect MMP-9 and uPA in 67 cases of gastric cancer. Results 1. Of the 67 cases gastric cancer, 34(50 75%) were positive for MMP-9, 35(52 24%) for uPA. The co-expression of MMP-9 and uPA was 26(38 .81%). 2. The MMP-9 positive rate in well differentiated carc inoma was higher than that in poorly one( P <0.05) ; The same result was concl uded about uPA( P <0.01). 3. MMP-9 and uPA positive rates in the cases with l ymph node metastasis were relatively higher than that without lymph node metasta sis( P <0.01). 4. The differences of MMP-9 and uPA positive rates were signif icant among cases of early gastric cancer and cases with muscularis involvement and cases with whole layer infiltration( P <0.01). 5. The relationship was sig nificant between the expression of MMP-9 and uPA in gastric cancer( P <0.01). Conclusion The study shows that MMP-9 and uPA may play an impo rtant role in histologic types or invasion or metastasis of gastric cancer. The assay of MMP-9 and uPA in patients with gastric cancer, as the clinical valuabl e marks, has the active role and instructive significance in estimating prognosi s and work out rational therapy.
