摘要
目的 :测定木瓜凝乳蛋白酶及其与 5 氟尿嘧啶 (5 Fu)、氨甲喋呤 (MTX )单独或联合作用对人肝癌细胞740 2的细胞毒性。方法 :采用MTT法 ,不同浓度的酶和药物在不同的培养时间下 ,通过其细胞抑制率和半数抑制率 (IC50 )测定木瓜凝乳蛋白酶及联合药物的细胞毒性。结果 :木瓜凝乳蛋白酶对 740 2细胞的IC50 约为 12 5 μg/mL ,随浓度增加对细胞的抑制率增加。培养 3 6h时酶对细胞的毒性最大。酶与 5 Fu、MTX联合使用比单独使用效果更好。结论 :木瓜凝乳蛋白酶对人肝癌细胞 740 2具有细胞毒作用 ,酶在培养 3 6h时细胞毒作用最大 ;
Objective To investigate the cytotoxicity of chymopapain and combining with 5 FU or MTX to the human hepatoma cell line 7402 cultured in vitro . Methods MTT method was employed in the assay to detect cytotoxicity of chymopapain to human hepatoma cell 7402 cultured in different drug concentrations and different times in vitro . Results Chymopapain showed cytotoxicity to human hepatoma cell 7402 (IC 50 is about 125 μg/mL) and had a concentration dependency. The chymopa pain had a maximum cytotoxicity when it had been cultured to 36 hours. Cooperation effect was showed when chymopapain combined with 5 FU or MTX to hepatoma cell line 7402. Conclusion Chymopapain has cytotoxicity to human hepatoma cell line7402 cultured in vitro . The maximum effect was displayed at the 36th hou after chymopapain was added. Cooperation effects were showed when chymopapain combined with 5 FU or MTX.
