摘要 研究结果表明,独蒜兰组织培养最好选用种子萌发的幼芽体进行。激素的种类和浓度影响愈伤组织的形成,在2,4 D2mg·L-1+6 BA0 25mg·L-1和2,4 D2mg·L-1+KT0 25mg·L-1的MS培养基中,外植体的死亡率较低;高浓度的2,4 D有利于快速形成大量的愈伤组织,但愈伤组织褐变死亡率较高;无论是种子萌发的芽还是从假鳞茎上剥下的芽,在6 BA2mg·L-1的MS上其长势都是最好;6 BA2mg·L-1最适合外植体为种子萌发的芽的增殖,当6 BA浓度为4mg·L-1时,则抑制了外植体的芽的增殖与生长;NAA对外植体的芽的增殖和生长都有抑制作用;MS+6 BA3 0mg·L-1+NAA0 5mg·L-1培养基对不同外植体芽的增殖倍数有显著性差异;添加了IBA的培养基有利于根系的诱导。 Based on the study,the juvenile buds germinated from seed of common pleione is the best material as exophytic using for tissue culture. The types of hormone and its concentration are the main affecting factors for formation of wound callus. The death rate of explants, which cultured on the medium of MS+2,4-D 2 mg·L^(-1)+6-BA 0.25 mg·L^(-1) and MS+2,4-D 2 mg·L^(-1)+KT 0.25 mg·L^(-1), is relatively low compared to others. High concentration of 2,4-D is advantageous to quick formation of more wound callus, but the death rate of wound callus is higher because of browning. Both of juvenile buds from seeds and pseudobulbs are all growing well on the medium of MS+6-BA 2 mg·L^(-1); and 6-BA 2 mg·L^(-1) is more suitable for multiplication of buds which from seeds and used as explants. But when the concentration of 6-BA is higher as 4 mg·L^(-1), growing and multiplication of exophytic buds were inhibited, and NAA has inhibition effect on growing and multiplication of multiplication as well. The medium of MS+6-BA 3.0 mg·L^(-1) + NAA 0.5 mg·L^(-1)showed significantly different effects on multiplication times to various explants,adding IBA is favor to derive roots.
