特异种质烟草全长cDNA文库的构建及质量分析 被引量:15

Construction of Full-length cDNA Library from A NativeTobacco Germplasm and Its Quality Analysis

摘要 采用SMART(switchingmechanismat5′endofRNAtranscript)技术构建了我国特有地方烟草品种HZNH的全长cDNA基因文库。大肠杆菌XL1-Blue平皿测定和PCR快速鉴定表明,文库的库容量为2 47×106pfu·mL-1,重组率为98 4%,所构建的文库达到了用于目的基因的分离筛选和克隆表达的建库要求。 To study the molecular mechanism of resistance to diseases and adversity environment of a native tobacco germplasm HZNH, and obtain their resistance genes, full-length cDNA library of HZNH was constructed by using SMART (switching mechanism at 5′end of RNA transcript) technology. According to the results of phage plaques bright selection of host bacterial strain XL1-Blue,PCR detection, the percentage of recombinant phages were 98.4% and the titer was 2.47×10~6 pfu·mL^(-1),which indicated that the library was of high quality for cloning target genes and expressing target proteins.
