Effects of HIF-2α and VEGF on Angiogenesis and Metastasis of Renal Cell Carcinoma

摘要 目的 探讨缺氧诱导因子 -2α(Hypoxiainduciblefactor -2α ,HIF -2α)、血管内皮生长因子 (vascularendothelialgrowthfac tor ,VEGF)在肾细胞癌中的表达及其与血管生成、癌转移的关系。方法 应用免疫组织化学技术检测 48例肾细胞癌中HIF -2α、VEGF的表达。结果  48例肾细胞癌HIF -2α阳性率为 70 8%,强表达 12例 ,中度表达 10例 ,弱表达 12例 ,无表达 14例 ,与正常对照组比较有显著性差异 (P <0 0 1)。Ⅰ级、Ⅱ级、Ⅲ~Ⅳ级VEGF阳性细胞数三组比较有显著性差异 (P <0 0 1)。患者癌转移者与无癌转移者HIF -2α、VEGF比较有显著性差异 ( P <0 0 1) ,HIF -2α表达与肾细胞癌恶性程度呈正相关 (r =0 682 ,P <0 0 1)。VEGF与HIF -2α表达呈正相关 (r =0 72 2 ,P <0 0 1)。结论 HIF -2α与VEGF有相关性 ,HIF -2α、VEGF与血管生成、癌转移有关 ,HIF Objective To explore the expression of HIF-2α and VEGF in renal cell carcinoma (RCC) and their correlation with angiogenesis and metastasis of renal cell carcinoma. Methods Expression of HIF-2α and VEGF were assessed by immunohistochemical straining in 48 cases of renal cell carcinomas. Results In 48 cases of renal cell carcinomas, positive rate of HIF-2α was 70.8%. Strong expression of HIF-2α was observed in 12 cases, moderate in 10 cases, weak in 12 cases and no expression in 14cases.There was a significant difference in HIF-2α expression between carcinoma tissues and control tissues (P<0.01). Every grade of RCC expressed VEGF and there were significant differeces in positive cell counts of VEGF among grades Ⅰ,Ⅱ and Ⅲ~Ⅳ of RCC (P<0.01). The expression of HIF-2α was positive relation with pathological grade of renal cell carcinoma (r=0.682,P<0.01). The expression level of HIF-2α and VEGF was significantly higher in metastatic RCC than that in no metastatic RCC. There was a positive correlation between HIF-2α and VEGF expression (r=0.722,P<0.01). Conclusions VEGF and HIF-2α expression highly correlated with angiogenesis and metastasis of RCC. The detection of VEGF and HIF-2α may serve as an index for discriminating potential tumor metastasis and prognosis in RCC.
