摘要
目的 :探讨端粒酶抑制剂对动物在体肿瘤的治疗作用及对放射治疗的增敏价值。方法 :将 SGC- 790 1胃癌细胞接种 BAL B/ c小鼠 ,采用端粒酶抑制剂 AZT联合放疗治疗小鼠胃癌 ,观察其对肿瘤体积和肿瘤端粒酶活性的影响。结果 :未治疗组小鼠肿瘤体积增加了 2 .5倍 ,而 AZT组、放疗组及 AZT联合放疗组肿瘤体积分别缩小了 2 6 .3%、36 .8%及 70 .5 %。肿瘤端粒酶活性检测 ,未治疗组、AZT组、放疗组及 AZT联合放疗组的相对端粒酶活性分别为 10 0、5 5 .2、2 4 .5及 12 .1。结论 :AZT、放疗均能使肿瘤体积缩小并降低端粒酶活性 ,且 AZT能增加肿瘤的放射敏感性。
Objective:To investigate the treatment effects of telomerase inhibitor for animal tumor in vivo and the radiosensitive value of telomerase inhibitor. Methods:The telomerase inhibitor AZT and radiotherapy were used to treat gastric cancers(SGC-7901) xenograft in BALB/c mice .The tumor was weekly measured and telomerase activity was examined by Semi-TRAP assay. Results:The tumor size of mice increased 3.3 times in untreated group. AZT, radiotherapy and AZT combined with radiotherapy reduced tumor volume of 26.3%,36.8% and 70.5%.The relative telomerase activity was 100 in untreated group,55.2 in AZT, 24.5 in radiotherapy, and 12.1 in AZT combined with radiotherapy group. Conclusion:Both AZT and radiotherapy are effective to treat murine gastric cancer though decreasing telomerase activity and reducing tumor size. AZT can increase the radiation sensitivity for murine gastric cancer.
