摘要
目的 :建立西洋参中人参皂苷Re ,Rb1的胶束电动毛细管色谱测定方法 ,考察影响分离的各因素 ,确定最佳色谱条件。方法 :测定缓冲液为含 2 0mmol·L-1硼酸、2 0mmol·L-1硼砂、6 0mmol·L-1胆酸钠和 2 0 %乙腈的混合溶液。电压 2 0kV ,柱温 2 5℃ ,检测波长 2 0 3nm ,压力进样 5s。结果 :线性范围Re为 0 .38~ 1.6 5mmol·L-1,Rb1为 0 .4 2~ 1.76mmol·L-1。加样回收率Re为 97.2 % ,RSD =1.6 % ;Rb1为 97.7% ,RSD =1.9% (n =5 )。结论 :该方法具有样品前处理简单 ,重现性好 ,所得色谱图信息丰富等特点。
Objective: A Micellar electrokinetic chromatography (MEKC) technique for the determination of ginsenosides Re, Rb_1 in Panax quinquefolius was developed and validated. Method: The MEKC was performed in a mixed buffer solution containing 20 mmol·L -1 boric acid, 20 mmol·L -1 sodium tetraborate, 60 mmol·L -1 sodium cholate (CA) and 20% acetonitrile under the applied voltage of 20 kV at 25 ℃. The detection wavelenth was 203 nm, the sampling time is 5 sec (hydrostatic injection). Result and Conclusion: The liner range was 0.38~1.65 mg·ml -1 for Re and 0.42~1.76 g·L -1 for Rb_1. The average recovery for Re was 97.2%, RSD=1.6% and that for Rb_1 was 97.7%, RSD=1.9% ( n =5). The preparation of sample is easy and the chromatogram has much information.
