摘要
利用顺铂(DDP)腹腔注射制备化疗大鼠食欲不振模型。方法:观察每只大鼠的进食量、体重、粪便等情况4天,之后大鼠腹腔注射DDP(0.5 g/L)5 mg/kg1一次,连续观察5 d,第5天再次注射DDP5 mg/kg1。结果:DDP注射的当日大鼠即出现进食量的明显下降,以后继续下降,部分大鼠甚至出现拒食,在化疗后第4天食量开始恢复。检测发现模型组体重下降、胃粘膜有损伤,胃肠粘膜有炎症,十二指肠5-羟色胺(5-HT)明显增高。结论:应用DDP可造成大鼠食欲不振模型。
To establish the rat model of Chemotherapy-induced anorexia (CIA) with cisplatin (DDP), the food consumption ,the body weight and stool had been observed for 4 days. Then the rats were injected ip DDP(0.5mg/ml)5mg·kg-1 at the fifth day, thereafter the rats were in-jected DDP once again. The body weight, the gastric mucosal lesion score, gastrointestinal tract hematoxylin-eosion staining and 5-hydroxytryptamine (5-HT) expression were also determined. The rat's food consumption was decreasing immediately from the first day of injection DDP, and some of rats even ate nothing. But at the fourth day the food consumption began to increase. we found there were gastric mucosal lesion, weight loss, inflammation in gastrointestinal tract, rise of 5-HT expression in model group. It was suggested that the rat model of CIA can be established by using DDP.
