表皮生长因子对大鼠垂体瘤催乳素分泌的影响 被引量:1

摘要 目的 采用乙烯雌酚 (DES)诱发垂体催乳素瘤模型 ,观察表皮生长因子在垂体催乳素瘤发生发展过程中的作用及其机制 ,为探明催乳素瘤的产生机制提供佐证。方法 采用Wistar系雌性大鼠 ,切除双侧卵巢后皮下埋植DES诱发垂体增生 ,检测DES诱发不同时期大鼠垂体前叶重量和血清催乳素水平 ,放免法检测原代培养大鼠垂体前叶细胞分泌的催乳素。结果 DES诱发的大鼠垂体前叶重量明显高于对照组 ,血清中催乳素含量明显增加 ,表皮生长因子使体外培养正常大鼠垂体前叶细胞催乳素的分泌增加 ,对乙烯雌酚诱发的大鼠垂体瘤催乳素的分泌没有显著作用。结论 表皮生长因子对乙烯雌酚诱发的大鼠垂体瘤催乳素的分泌没有显著性的影响 ,催乳素瘤细胞对表皮生长因子的反应性下降。 Objective To investigate the role and the effect of epidermal growth factor(EGF) on the secretion of prolactin in the course of formation of pituitary prolactioma induced by diethylstilbestrol. Methods The pituitary prolactinoma was induced by implanting subcutaneous diethylstibestrol in ovariectomised Wistar rats. Adenohypophysis was weighted, and prolactin levels in serum and primary cultured hyperplasia adenohypophysis cells of rat were measured by radioimmunoassay. Results Both adenohypohysis weights and levels of serum prolaction in prolactinoma rat were obviously higher than those in control group. EGF enhanced the secretion of prolactin in primary cultured adenohypophysis cells of normal rats but had no influence over prolaction secretion in diethylstilbestrol induced prolactinoma cells in vitro.Conclusion EGF had no significant influence over prolactin secretion in Diethylstilbestrol-induced prolactinoma cells, the reaction of prolactinoma cells to EGF was declined.
