三倍体毛白杨组培快繁和工厂化育苗技术研究

Study on in vitro fast-propagation and industrialization breeding techniques of triploid Chinese white poplar

采用1年生三倍体毛白杨优质苗木单芽茎段,研究其组织培养技术和工厂化育苗技术.研究结果表明,组培苗增殖与生根同步进行,通用最佳培养基为:H+NAA0.15mg/L,生根率80%～98%,年增殖次数8次,繁殖系数为5.蛭石粉+泥炭土 1∶1 是三倍体毛白杨组培苗炼苗的最佳基质.通过组培苗生产、温棚炼苗和大田炼苗试验,研究出了包括接种培养阶段、移栽和苗圃管理阶段3个连续过程的三倍体毛白杨工厂化育苗的工艺流程.

The tissue culture and industrialization breeding techniques were studied by adopting the stem segment of single bud of Populus tomentosa one-year-growth seedling. The study results indicated that the proliferation and radication of in vitro seedling were made simultaneously, the common-used culture medium was H+NAA 0.15 mg/L and radication rate was 80%~98%. And it proliferated 8 times annually and its propagation index was 5; The medium of vermiculite + turf with 1∶1 proportion was best for transplanting and training of Populus tomentosa in vitro seedling. Through production of in vitro seedling and trial of seedling planting in warm shed and open field, craft process of industrialization breeding of Populus tomentosa was put forward in its 3 continuous courses of inoculation, cultivation, transplant and nursery garden management stages in this paper.
