RAPD analysis of some core germplasms in tobacco

摘要 本研究对24个不同类型烟草核心种质进行了RAPD标记。通过43个RAPD引物PCR扩增的214个分子量不同的DNA片段,标记出24个烟草核心种质的指纹图谱,表现了高度的遗传多样性。通过聚类分析,将24个种质分为6大聚类群,类群Ⅰ包含11种质,其中烤烟5个,晒烟3个,白肋烟2个,雪茄烟1个;类群Ⅱ包含9个种质,其中烤烟2个,晒烟2个,白肋烟2个,香料烟2个,马里兰烟1个;类群Ⅲ、Ⅴ,分别包含晒烟、黄花烟各1个;Ⅳ、Ⅵ为2个野生种。各类群遗传差异由大到小顺序为Ⅵ>Ⅴ>Ⅳ>Ⅲ>Ⅱ>Ⅰ。种质间的遗传差异并不完全取决于栽培类型间的差异,而与种质间演化关系有关,野生种与栽培种间存在较大的遗传差异。 This research carried out RAPD markers for 24 different type tobacco core germplasms. With 43 RAPD primers, the 214 DNA segments of different molecular weigh were PCR amplified, marked the fingerprint pattern of 24 tobacco core germplasms. Through clustering, 24 germplasms were divided into 6 clusters, I contains 11 germplasms, in which 5 flue-cured tobacco, 3 sun cured tobacco, 2 hurley tobacco and 1 cigar. Cluster Ⅱ contains 9 germplasms, 2 flue-cured tobacco., sun cured tobacco、hurley and oriental tobacco, and 1 for Maryland tobacco. Cluster Ⅲ , V each containd 1 sun cured tobacco and 1 N. rustica, Ⅳ and Ⅵ had only 2 wild species. The genetic difference of all clusters were ordered from larger to small as Ⅵ>V>Ⅳ>Ⅲ>Ⅱ> I . The genetic difference between germplasm depend on mainly their evolution relation, and not related consequentially to their cultivar type. Between species and cultivar, existed greater genetic difference.
