摘要
目的 观察环磷酰胺对人喉鳞状细胞癌Hep 2细胞端粒酶活性及其p16表达的影响 ,以揭示环磷酰胺的抗癌机制。方法 运用体外细胞培养、端粒酶重复序列扩增 酶联免疫吸附法 (TRAP ELISA)及流式细胞技术观察、分析不同浓度的环磷酰胺作用 72h的Hep 2细胞。 结果 环磷酰胺能以浓度依赖方式下调端粒酶活性并诱导细胞 p16基因的表达增强。端粒酶活性下调与p16基因表达紧密相关。 结论 环磷酰胺能下调端粒酶活性、诱导细胞增强p16的表达 。
ObjectiveTo investigate effect on telom erase activity and p16 gene expression of human laryngeal carcinoma cells Hep-2 induced by cytoxan (CTX). MethodsBy techniques of cell culture in vitro, telomeric repeat amplification protocol with ELISA (TRAP-ELISA) and flow-cytometry (FCM), Hep-2 cell was treated by cytoxan in various concentrations for 72 h. ResultsCytoxan down-regulated telomerase activity of Hep-2 cell and increased expression of p16 gene protein in a concentration-dependent manner. There was a close correlation between telomerase activity and the expression of p16 gene protein. ConclusionCytoxan could down-regulate telomerase activity and induce expression of p16 gene protein, which may be one of important mechanisms of cytoxan's anticancer action.
