摘要
目的 :探讨黄芪对实验性病毒性心肌炎细胞凋亡及 bcl- 2 / bax基因的影响。方法 :Balb/ c小鼠 2 4只腹腔感染柯萨奇病毒 B3(CVB3)后随机分成 2组 :黄芪治疗组 (每天腹腔注射黄芪注射液 1 0 g/ kg)及对照组 ,第 8天取心脏标本 ,进行病理学检查、细胞凋亡 TUNEL 法检测以及逆转录聚合酶链反应 (RT- PCR)分析 bcl- 2和 bax m RNA的表达水平。结果 :1黄芪治疗组病变积分明显低于对照组 (P <0 .0 1 ) ;2黄芪治疗组心肌细胞凋亡指数 (AI)明显低于对照组 (P <0 .0 1 ) ;3黄芪治疗组心肌组织 bcl- 2的 m RNA的相对表达量明显高于对照组 (P <0 .0 1 ) ,bax m RNA的相对表达量无明显改变 (P >0 .0 5 ) ,bcl- 2m RNA/ bax m RNA比值则明显增高 (P <0 .0 5 )。结论 :黄芪可通过上调病毒性心肌炎小鼠心肌组织 bcl- 2的基因转录 ,抑制细胞凋亡 。
Objective:To investigate the effects on apoptosis and bcl 2/bax gene transcription in development of virus myocarditis in mice treated with astragalus.Methods:Virus myocarditis model was created by intraperitoneal inoculation with coxsackievirus B3 solution and were divided into astragalus group and control group. The myocardial pathologic changes were observed, and apoptosis of cardiomyocyte and levels of Fas/FasL mRNA were detected respectively by insitu end labeled DNA(TUNEL) and reverse transcription polymerase chain reaction(RT PCR).Result:①Pathology scores in astragalus group was significantly lower than that in control group(P<0 01).②Apoptotic index in astragalus group was significantly lower than that in control group(P<0 01).③The mRNA content of bcl 2 in astragalus group was significantly higher than that in control group(P<0 01),bax mRNA content was unchanged, and bcl 2 mRNA/ bax mRNA was also significantly higher(P<0 05).Conclusion:Gene transcription of bcl 2 may be up regulated in the development virus myocarditis in mice treated with astragalus. The change may contribute to reduction of the apoptosis of cardiomyocyte and myocardial injury.
