摘要
目的 :探讨玉郎伞提取物 (YL S)对四氯化碳 (CCl4 )、硫代乙酰胺 (Thioacetamide,TAA)和对乙酰氨基酚(Acetam inophen,AP)致小鼠急性化学性肝损伤的保护作用及其机制。方法 :采用 CCl4 、TAA和 AP所致的实验性肝损伤模型 ,通过测定 AL T、肝药酶和丙二醛 (MDA)、谷胱甘肽 (GSH)、谷胱甘肽过氧化物酶 (GSH- PX)等的改变 ,观察 YL S对急性化学性肝损伤的影响。结果 :YL S对 CCl4 、TAA和 AP所致急性肝损伤小鼠有明显的降低 AL T活性 ,抑制脂质过氧化、降低肝组织 MDA含量 ,提高肝脏细胞色素 P4 5 0 (P4 5 0 )和细胞色素 b5 (Cyt.b5)含量 ,增强苯胺羟化酶和氨基比林 - N-去甲基酶活性 ,以及诱导肝药酶活性作用。此外 ,YL S还能明显提高 AP肝损伤小鼠肝组织和血清中谷胱甘肽 (GSH)及谷胱甘肽过氧化物酶(GSH- PX)的水平。结论 :YL S对小鼠急性化学性肝损伤具有显著的保护作用 ,其机制可能与抑制脂质过氧化、诱导肝药酶、提高 GSH含量和 GSH-
Objective:To study the protective effects and mechanisms of Yulangsan extract (YLS) on CCl 4, thioacetamide (TAA) and Acetaminophen(AP) induced acute liver injuries in mice.Methods:The models of CCl 4, TAA and AP induced acute liver injury in mice were used to study the effects of YLS on acute liver injury.Result:YLS could significantly decrease the elevation of serum transaminase (ALT)in liver injuries induced by CCl 4,TAA and AP in mice, increase the contents of both hepatic cytochrome P450 (P450) and cytochrome (Cyt.b5), and the activities of hepatic microsomal aniline hydroxylase(AH) and aminopyrine N demethylase(AND).YLS could inhibit lipid peroxidation and the formation of MDA . YLS could also markedly increase the activity of GSH PX and the content of GSH in both serum and liver respectively.Conclusion:YLS has protective effects against CCl 4 ,TAA and AP induced acute liver injuries in mice, possibly due to its inhibition of lipid peroxidation, induction of hepatic microsomal drug metabolizing enzyme, and the increase in the activity of GSH PX and the content of GSH.
